摘要
目的观察异甘草素对大鼠脑胶质瘤组织细胞间黏附分子1(ICAM-1)和血管细胞黏附分子-1(VCAM-1)表达的影响,探讨异甘草素抗胶质瘤作用机制。方法清洁级Wistar雌性大鼠,n=36只,随机分为:假手术组(n=12),C6胶质瘤模型组(n=12)与异甘草素组(n=12)。异甘草素组腹腔注射异甘草素,500μg/kg/d,模型组腹腔注射等量生理盐水,连续给药一周后处死大鼠。免疫组织化学方法和Western blot方法检测肿瘤组织ICAM-1和VCAM=1蛋白表达水平的变化。结果免疫组织化学和Westem blot结果显示,与假手术组相比,模型组与异甘草素组大鼠脑组织ICAM-1和VCAM-1的蛋白表达的平均光密度值显著增加(P<0.01)。两两比较,异甘草素组大鼠脑组织ICAM-1和VCAM-1的蛋白表达的平均光密度值显著低于模型组(P<0.01)。结论异甘草素抑制胶质瘤的作用可能与降低脑胶质瘤大鼠ICAM-1和VCAM-1的表达相关。
Objective To investigate the effect of isoliquiritigenin on the expressions of intercellular adhesion molecule-l(ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in C6 glioma rat models and to explore the antiglioma mechanism. Methods 36 Wistar female rats were randomly divided into 2 group: sham group(n=12)and C6 glioma model group(n=24) and isoliquiritigenin group(n=12). The rats of isoliquiritigenin group were treated by intraperitoneal injection of isoliquiritigenin(500 μ g / kg / d), and the rats of model group intraperitoneal injection of normal saline. All rats were executed one week after continuous medicine.The expressions of ICAM-1 and VCAM-1 were detected by immunohistochemistry and western blot methods. Results The mean optic density(MOD) values of ICAM-1 and VCAM-! positive expressions in brain tissues were higher significantly in model group than in sham group(P〈0.01), but lower significantly in isoliquiritigenin group than in the model group(P〈0.01). Conclusion The inhibition effect of Isoliquiritigenin on glioma might be related to the downregulation of ICAM-1 and VCAM-1 expression levels in brain tissue of rats with brain glioma.
