破血逐瘀中药虻虫对荷H22肝癌小鼠免疫功能及细胞凋亡的影响 被引量:8

摘要 目的:观察破血逐瘀中药虻虫对荷H22小鼠肝癌病理、瘤重、脾重、IL-2及凋亡指数的影响。方法:雄性KM小鼠50只随机分为模型,化疗组,虻虫高、中、低剂量组。腋下注射H22细胞建立荷H22肝癌小鼠模型。虻虫治疗组以0.045、0.09、0.18g·kg^(-1)·d^(-1)剂量灌胃给药,化疗组5-FU以25mg·kg^(-1)·d^(-1)剂量腹腔注射给药,隔日一次;模型组予以等体积生理盐水灌胃给药2w后处死小鼠,称取瘤重脾重,计算瘤指数及脾指数,ELISA法检测血清IL-2水平,TUNEL法检测肝癌细胞凋亡,HE染色观察肝癌组织形态变化。结果:高、中剂量虻虫均可使肝癌组织呈大片坏死,并可明显降低瘤指数及提高IL-2水平及凋亡指数,与模型组比较,有显著性差异(P<0.01);高剂量虻虫可明显升高脾指数,与模型组比较,有显著性差异(P<0.01)。结论:破血逐瘀中药虻虫能显著抑制荷H22小鼠肝癌细胞增殖,其机理可能与促进肝癌细胞坏死,降低瘤指数,升高脾指数及血清IL-2水平,从而增强机体免疫及促进肝癌细胞凋亡有关。 Objective :To observe the influence of breaking stagnant and eliminating blood stasis medicine gadfly on Pathology, tumor weight, spleen weight, IL -2 and apoptosis index in H22 tumor - bearing mice. Methods: 50 male KM mice were randomly divided into model group, chemotherapy group and gadfly of high, medium and low dose group. The establishment of H22 tumor model of axillary injection of H22 cells. The treatment group of gadfly with 0. 045,0.09,0. 18g·kg^-1·d^-1 dose to intragastric administration, chemotherapy group 5 - FU with 25 mg·kg^-1·d^-1 dose to intragastric administration, injected 1 times alternate - day. Model group were given saline to gavage. Mice were killed after administration of 2W. The tumor and spleen were weighed to calculate the tumor index and spleen index. The serum level of IL - 2 was detected by ELISA method. Apoptosis was detected by TUNEL assay. The morphological changes of cells were observed by HE mothed. Results : High and middle dose of gadfly would make liver cancer tissue showed large areas of necrosis,and would significantly reduce tumor index and improve IL - 2 and apoptosis index,there was a significant difference compared with model group(P 〈0.01) ; High dose of gadfly would significantly increase the spleen index , there was a significant difference compared with model group( P 〈 0.01). Conclusion : Breaking stagnant and eliminating blood stasis medicine Gadfly can significantly inhibit proliferation of the mice bearing H22 hepatoma cell and the mechanism may be connected with promote liver cell necrosis, decrease the tumor index and increase the level of IL -2 in serum, thereby enhance the immunity function and promote apoptosis of hepatoma cells.
