隔附子饼灸对肠易激综合征大鼠内脏高敏感的机理研究

The Mechanisms of Herbs- partition Moxibustion Treating Visceral Hypersensitivity in Rats with Irritable Bowel Syndrome is through Brain- gut Interaction Axis

目的:探讨隔附饼灸对肠易激综合征大鼠高敏感性的机理。方法:本实验参照AL-chaer ED等的方法制备IBS-D大鼠模型,随机分成正常组、模型组、隔药饼灸组和匹维溴铵组,每组8只。采用不同处理方法后,进行AWR评分,用qRT-PCR检测各组结肠、脊髓和下丘脑的orexin mRNA表达。结果:与正常组比较,各级压力下模型组AWR评分明显升高(均P﹤0.01),与模型组相比,隔药饼灸组和匹维溴铵组AWR评分明显降低,差异存在统计学意义(均P﹤0.01);与正常组比较,模型组大鼠的结肠、下丘脑、脊髓Orexin A mRNA表达均明显降低,差异存在着统计学意义(均P﹤0.05),与模型组比较,隔药饼灸组、匹维溴铵组大鼠的结肠、下丘脑、脊髓Orexin A mRNA表达明显增强,差异存在着统计学意义(均P﹤0.05)。结论:隔药饼灸治疗IBS的机理可能是通过脑肠互动轴调节CNS系统、ENS系统增加orexin的表达,降低内脏高敏感而提高大鼠的痛阈。

Objective ： To explore the mechanisms of herbs - partition moxibustion on brain - gut interaction axis in rats with irritable bowel syndrome. Methods ： The model of IBS were established by Method AL - chaer E D. 36 rats were randomly divided into model group, herbs - partition moxibustion group and pinaverium bromide group, each groups has 8rats, 8 rats were as normal group. After different processing methods, AWR scores were measured and orexin. mRNA expression of colon, spinal cord and hypothalamic were detected by qRT - PCR. Results ： Compared with normal group, AWR scores in other groups under all kinds of pressures were significantly increased （ all P 〈 0. 01）. However, compared with model group, AWR scores of herbs - partition moxibustion group and pinaverium bromide group under all kinds of pressure were significantly decreased （al l P〈0.01）. compared with normal group, ex-pression of mRNA in colon, spinal cord and hypothalamus were decreased （ all P 〈0.01） , compared with model group, expression of mRNA in colon, spinal cord and hypothalamus were significantly increased （ all P 〈0.01） . Conclusions： Mechanism of herbs - partition moxibustion for treating IBS is that it can increase the expression of Orexin and decrease visceral hyposensitivity to raise the pain threshold by adjusting CNS system and the ENS system through brain - gut interaction axis.
