摘要 采用饱和硫酸铵盐析法和低温乙醇法,对牦牛血清免疫球蛋白进行分离提取,比较两种方法的提取得率和产品回收率,结果表明采用低温乙醇法提取效果最佳,最佳提取条件:蒸馏水稀释倍数为3倍,乙醇添加量为25%,NaCl溶液的浓度为0.10mol·L^(-1),pH5.0。其提取得率为9.205mg/ml,产品回收率为89%,是适合于血清中免疫球蛋白粗提的有效方法。 Immunoglobulin is a kind of important immunologic active material in living body. It plays a very im-portant role in adjusting the function of various living organs and systems. In this paper, immunoglobulin from yak ser-um was isolated by saturated (NH4)2S04 salting - out method and the low - temperature ethanol method, and obtaining crude extract immunoglobulin. The extraction rate and product's recovery rate by two methods were compared. The results showed that the better method is low - temperature ethanol method, with its extraction percentage being 9. 205mg/mL and product's recovery rate being 89% . The optimal extraction method : the distilled water dilution multiple is 3times, the additive amount of ethanol is 25% , the concentration of NaCl solution is 0. 10 mol· L-1, and the pH value is 5. 0.
