鱼腥蓝细菌实时观察微型培养系统的建立和应用

摘要 为克服传统方法不能实现对特定菌丝或细胞的连续定点观察的难题,利用低熔点琼脂糖培养基将菌丝包埋在玻底培养皿中,制备了鱼腥蓝细菌PCC 7120菌株实时观察的微型培养体系。结果表明:在该培养体系中,菌丝能进行正常的生长、分裂和分化;利用该系统可以观察细胞分裂蛋白FtsZ和DNA双链损伤修复蛋白RecN在细胞分裂过程中的动态变化,实现对同一菌丝进行长时间的连续定点观察。 The filaments were embedded in glass bottom plate to fix filaments by using low melting point agarose. A real-time observation of microscopic culture system for Anabaena sp. strain PCC 7120 was prepared. Results showed that Anabaena sp. strain PCC 7120 grew well in glass bottom culture dish,and exhibited normal activity of cell division and differentiation. The continuous dynamics changes of cell division protein FtsZ and DNA double-stranded damage response of protein RecN during cell division process through this cultivation method were observed. It is indicated that continuous fixed-point observation of the same filament was successfully achieved.
