摘要
目的观察早期糖尿病肾病大鼠肾脏线粒体Ca2+及细胞色素C(Cyt C)的变化。方法将20只Wistar大鼠随机分为对照组、模型组,每组10只。模型组建立链脲佐菌素诱导早期糖尿病肾病大鼠模型,对照组大鼠未经链脲佐菌素诱导。第10周末测定两组血糖、肾质量指数、24h尿蛋白定量;检测两组大鼠肾脏线粒体Ca2+、Cyt C含量。结果与对照组相比,模型组大鼠的血糖、肾质量指数、24h尿蛋白定量均显著升高(均P<0.01),线粒体Ca2+及胞浆Cyt C含量升高,线粒体Cyt C含量显著降低(P<0.05)。结论早期糖尿病肾病大鼠肾脏线粒体内钙超载,线粒体Cyt C释放增加。
Objective To study the changes of renal mitochondrial calcium and cytochrome C levels in rats with early diabetic nephropathy(DN). Methods Model group was induced by injection of streptozotocin. Twenty male Wistar rats were divided into as control group and model group, with ten rats in each group. Ten weeks later, blood glucose level, renal mass index, 24 hour urinary protein excretion and levels of renal mitochondrial calcium and cytochrome C were measured. Results Compared with control group, Blood glucose level, renal mass index,24 hour urinary protein excretion , renal mitoehondrial calcium level were significantly increased (all P〈0.01) , cytoplasm cytochrome C levels increased and renal mitochondrial eytochrome C levels decreased in model group (P〈0.05). Conclusion This study suggested mitochondrial calcium overload and the release of mitochondriat cytochrome C were elevated in early DN rats.
