摘要
目的:研究氧化应激损伤对脊髓神经元细胞中miR-210表达的影响及miR-210抑制剂的神经保护效应。方法:培养大鼠背脊神经元细胞并分为对照组(A组)、H_2O_2组(B组)、NC组(C组)、H_2O_2+NC组(D组)、H_2O_2+miR-210抑制剂组(E组),测定A组、B组细胞中miR-210的表达量以及C组、D组、E组细胞的活力以及氧化应激分子、凋亡分子、功能相关分子的表达量。结果:B组脊髓神经元细胞中miR-210的表达量显著高于A组;处理12、18、24h,D组脊髓神经元细胞的OD值显著低于C组,E组脊髓神经元细胞的OD值显著高于D组;处理24h后,D组脊髓神经元细胞中HO-1、NOX-2、CAT、GAP-43、synapsin-I的含量均显著低于C组,ROS、8-OHdG、c-fos、c-jun、AP1、caspase-3、PSD-93、PSD-95的含量均显著高于C组;E组脊髓神经元细胞中HO-1、NOX-2、CAT、GAP-43、synapsin-I的含量均显著高于D组,ROS、8-OHdG、c-fos、c-jun、AP1、caspase-3、PSD-93、PSD-95的含量均显著低于D组。结论:氧化应激损伤会造成脊髓神经元细胞中miR-210高表达,miR-210抑制剂能够减轻氧化应激所造成的神经元损伤。
Objective:To study the effect of oxidative stress injury on miR-210 expression in spinal neurons and the neuroprotective effect of miR-210 inhibitor.Methods:Spinal neurons of rats were cultured and divided into control group(group A),H2O2group(group B),NC group(group C),H2O2+NC group(group D)and H2O2+miR-210 inhibitor group(group E),miR-210 expression levels in group A and group B as well as the cell vitality and expression levels of oxidative stress molecules,apoptotic molecules and function-related molecules in group C,group D and group E were determined.Results:miR-210 expression level in group B was significantly higher than that in group A;after 12 h,18hand 24 hof treatment,OD values ofgroup D were significantly lower than those of group C,and OD values of group E were significantly higher than those of group D;after 24 h of treatment,HO-1,NOX-2,CAT,GAP-43 and synapsin-I levels in group D were significantly lower than those in group C while ROS,8-OHdG,c-fos,c-jun,AP1,caspase-3,PSD-93 and PSD-95 levels were significantly higher than those in group C;HO-1,NOX-2,CAT,GAP-43 and synapsin-I levels in group E were significantly higher than those in group D while ROS,8-OHdG,c-fos,c-jun,AP1,caspase-3,PSD-93 and PSD-95 levels were significantly lower than those in group D.Conclusion:Oxidative stress injury can cause high miR-210 expression in spinal neurons,and miR-210 inhibitor can reduce the neuron injury caused by oxidative stress.
