摘要
目的:研究胶质瘤组织中miR-221表达量与PTEN/Akt信号通路、EMT相关基因表达的相关性。方法:收集在我院接受手术切除的35例高级别脑胶质瘤标本及瘤旁组织标本,抽提组织中的miRNA并测定miR-221的表达量,抽提组组中的蛋白并测定PTEN/Akt信号分子、EMT相关基因的表达量。结果:胶质瘤组织中miR-221的表达量以及PI3K、Akt、Ki67、N-cadherin、Vimentin、OPN、Snail、Slug的蛋白表达量显著高于瘤旁组织,PTEN、E-cadherin的蛋白表达量显著低于瘤旁组织样本;miR-221高表达脑胶质瘤组织中PTEN、E-cadherin的蛋白表达量显著低于miR-221低表达的脑胶质瘤组织,PI3K、Akt、Ki67、N-cadherin、Vimentin、OPN、Snail、Slug的蛋白表达量显著高于miR-221低表达的脑胶质瘤组织。结论:胶质瘤组织中高表达的miR-221能够靶向调节PTEN/Akt信号分子、EMT相关基因的表达,进而造成胶质瘤细胞增殖、侵袭。
Objective:To study the correlation of miR-221 expression level with PTEN/Akt signal pathway and EMTrelated gene expression in glioma tissue.Methods:A total of 35high-grade glioma samples and peritumoral tissue samples surgically removed in our hospital were collected,miRNA in tissue was extracted and the expression of miR-221 was determined,the protein in tissue was extracted and the expression of PTEN/Akt signaling molecules and EMT-related genes were determined.Results:miR-221 expression level as well as PI3 K,Akt,Ki67,N-cadherin,Vimentin,OPN,Snail and Slug protein expression levels in glioma tissue was significantly higher than those in peritumoral tissue while PTEN and E-cadherin protein expression levels were significantly lower than those in peritumoral tissue;PTEN and E-cadherin protein expression levels in glioma tissue with high miR-221 expression were significantly lower than those in glioma tissue with low miR-221 expression while PI3 K,Akt,Ki67,N-cadherin,Vimentin,OPN,Snail and Slug protein expression levels were significantly higher thanthose in glioma tissue with low miR-221 expression.Conclusion:Highly expressed miR-221 in glioma tissue can target and regulate the expression of PTEN/Akt signaling molecules and EMT-related genes,thus causing the glioma cell proliferation and invasion.
