摘要
目的观察甘露消毒丹及其残方对肠道病毒71型(EV71)感染后的RD细胞miRNA-155表达的影响。方法运用细胞培养技术,设甘全方组、清残方组、利残方组、利巴韦林组、正常细胞对照组、模型对照组,将药物作用于EV71感染后的RD细胞,24 h后RT-PCR法检测各组细胞miR-155表达。结果 1)模型对照组、甘全方组、清残方组、利巴韦林组miR-155表达均比正常细胞对照组低,利残方组比正常细胞对照组升高(P<0.05或P<0.01);甘全方组、清残方组、利残方组miR-155的表达比模型对照组高(P<0.05或P<0.01),利巴韦林组与模型对照组差异不明显(P>0.05);甘全方组、清残方组、利残方组miR-155表达比利巴韦林组升高(P<0.05或P<0.01),清残方组、利残方组的表达比甘全方组升高(P<0.05或P<0.01),利残方组最高。结论 EV71感染RD细胞后miR-155表达降低,与文献报道趋势不同,提示其表达可能具有细胞特异性;甘全方、清残方、利残方均能上调EV71感染后miR-155的表达从而调控免疫反应;利残方与清残方之间可能存在相互拮抗关系。
Objective: To observe Ganlu Xiaodu decoction and its incomplete prescriptions' effects on MiR- 155's expression in RD cells infected by EV71. Methods: With technique of cell cuhuring,Ganlu Xiaodu decoction therapy group ,incomplete Qing prescription therapy group ,incomplete Li prescription therapy group ,normal cells control group ,model control group and ribavirin control group were set,and then the expressions of miR-155 in these groups after intervention with drugs in 24 hours were detected by RT-PCR. Results: 1 )Compared with normal cells control group ,expression of miR-155 of model control group ,Ganlu Xiaodu decoction therapy group, incomplete Qing prescription therapy group and ribavirin control group all decreased,but that of incomplete Li prescription therapy group increased. 2)Levels of miR-155 in Ganlu Xiaodu decoction therapy group,incomplete Qing prescription therapy group and incomplete Li prescription therapy group were higher than model control group,but ribavirin control group had no significant difference with model control group. 3)Levels of miR-155 in Ganlu Xiaodu decoction therapy group, incomplete Qing prescription therapy group and incomplete Li prescription therapy group were higher than ribavirin control group,and levels of miR-155 in incomplete Qing prescription therapy group and incomplete Li prescription therapy group were higher than Ganlu Xiaodu decoction therapy group, and level of miR-155 in incomplete Li prescription therapy group was the highest. Conclusion: Expression of miR-155 in EVTl-infected RD cells decreases;the result is not consistent with what other literature reported, which indicates that expression of miR-155 may have cell specificity. Ganlu Xiaodu decoction,its incomplete Qing prescription and incomplete Li prescription all can regulate the immune reactions caused by infection of EV71 by increasing miR-155's expression. And there probably are antagonism effects between incomplete Qing prescription and incomplete Li prescription.
