转化生长因子-β诱导胃癌细胞人白细胞抗原-G表达机制的研究

摘要 目的 探讨转化生长因子-β诱导胃癌细胞人白细胞抗原-G的表达及可能作用机制。方法 胃癌患者20例,胃癌组织及癌旁组织标本,同时采集术前1天相应患者的外周血标本。肿瘤组织和以及癌旁组织在-80°液氮冷冻保存,10%福尔马林溶液中固定2-3天洗涤、浸腊、包埋以及防脱片。相应外周血标本中可溶性人白细胞抗原-G浓度检测。采用ELISA对转化生长因子-β和人白细胞抗原-G的检测,应用转化生长因子-βkit、可溶性人白细胞抗原-G kit检测胃癌细胞转化生长因子-β与人白细胞抗原-G的浓度。应用SPSS 17.0软件,所获数据采用皮尔逊相关分析法和2检验。结果 在胃癌细胞以及组织中转化生长因子-β及人白细胞抗原-G呈高表达水平,两者的表达呈相关性。不同浓度转化生长因子-β刺激能上调人白细胞抗原-G不同程度表达,转化生长因子-β表达和人白细胞抗原-G的表达在胃癌组织中及外周血中的浓度呈显著正相关。结论 转化生长因子-β参与了胃癌组织中人白细胞抗原-G的表达上调说明转化生长因子-β和人白细胞抗原-G在胃癌组织及外周血中的蛋白水平呈正相关;进一步阐明转化生长因子-β不仅与胃癌的活化和转移相关。 Objective To study the transforming growth factor- β person leucocyte antigen induced gastric cancer cells - G expression and the possible mechanism. Methods 20 patients with gastric cancer, gastric cancer tissue and tissue adjacent to carcinoma specimens, 1 day corresponding peripheral blood in patients with preoperative samples at the same time. Tumor tissue and tissue adjacent to carcinoma in - 80 o liquid nitrogen cryopreservation, fixed in 10% formalin solution 2-3 days washing, impregnated, embedding and overhand slice. Corresponding peripheral blood specimens of soluble leucocyte antigen - G concentration detection. By ELISA for transforming growth factor beta and human leukocyte antigen - G testing. Application of transforming growth factor- β kit, soluble human leukocyte antigen - G kit to detect gastric cancer cells transforming growth factor- β and the concentration of human leucocyte antigen - G. The application of SPSS 17.0 software. The data obtained using Pearson correlation analysis and χ2 test. Results In gastric cancer cells and tissues transforming growth factor- β and person leucocyte antigen - G has high exp ression level, both the expression of correlation. Different concentrations of transforming growth factor- β stimulation can raise leucocyte antigen - G express different level. Transforming growth factor beta expression and the expression of human leucocyte antigen - G in gastric cancer tissues and peripheral blood was significantly positively related to the concentration. Conclusion Transforming growth factor -β involved in gastric cancer tissue expression of human leucocyte antigen - G raised that transforming growth factor- β and human leukocyte antigen - G protein levels in gastric cancer tissue and peripheral blood were positively correlated. Growth factors further clarify- β is not only associated with the acti- vation and transfer of gastric cancer.
