维生素D3对小鼠实验性结肠炎和炎症相关结直肠癌的保护作用 被引量:4

摘要 目的探究预防性补充维生素D3(Vit D3)对小鼠急性实验性结肠炎(AC)和炎症相关结直肠癌(colitis-associated CRC,CAC)的保护作用及相关机制。方法采用葡聚糖硫酸钠(dextran sodium sulfate,DSS)诱导小鼠实验性结肠炎模型,氧化偶氮甲烷(AOM)联合DSS诱导CAC模型,干预组Vit D3定期灌胃。机制研究采用实时荧光定量PCR和免疫组化染色法。结果 Vit D3干预后,AC模型小鼠的结肠炎症减轻,其中结肠组织TNF-α表达和结肠上皮细胞凋亡减少。CAC模型小鼠的成瘤体积变小,其中β-catenin的表达水平和入核减少。结论预防性补充Vit D3对小鼠结肠炎症和CAC形成具有保护作用。 Objective To explore whether Vitamin D3 (VitD3) palys a protective role in acute experimental colitis (AC) and colitis-associated colorectal cancer (CAC) in mice and its mechanisms. Methods Dextran sodium sulfate (DSS) was used to induce AC mouse model. Azoxymethane and DSS were used to induce CAC model. VitD3 was given by gastric lavage. RT-PCR and immunohistochemieal (IHC) staining were used in mechanism studies. Results VitD3 reduced colitis severity in AC model in which colonic TNF-α expression and intestinal epithelial cell apoptosis were de- creased significantly after VitD3 intervention. VitD3 reduced tumor size significantly in CAC model, in which colonic β- catenin expression and nuclear localization were decreased significantly. Conclusion VitD3 palys a protective role in experimental colitis and CAC in mice.
