摘要
目的探讨乌司他丁是否可通过对NF-κB活性的抑制,起到治疗溃疡性结肠炎(ulcerative colitis,UC)的作用。方法 24只BALB/C小鼠以DSS法建立UC小鼠模型,造模后随机分为模型对照组、柳氮磺吡啶治疗组、柳氮磺吡+乌司他丁治疗组,每组8只,另8只正常小鼠设为正常对照组(饮用蒸馏水)。于用药后7 d对小鼠进行相关指标测定:疾病活动指数(DAI)、结肠组织病理学改变、血清中ALB、C反应蛋白、TNF-α的表达、结肠黏膜中SOD、NF-κB的表达。结果用药后各组小鼠DAI、C反应蛋白、TNF-α、NF-κB对比:正常对照组<柳氮磺吡啶+乌司他丁治疗组<柳氮磺吡啶治疗组<模型对照组;ALB、SOD的对比:正常对照组>柳氮磺吡啶+乌司他丁治疗组>柳氮磺吡啶治疗组>模型对照组。结论乌司他丁可明显抑制NF-κB的表达,对UC的治疗效果明显优于单纯使用柳氮磺吡啶。
Objective To explore whether Ulinastatin can inhibit the NF-KB activity and can exert effect on the treatment of ulcerative colitis (UC). Methods Twenty-four BALB/C mice were used to establish UC mice model in the method of DSS. After modeling, the mice were randomly divided into 3 groups, the model control group, Sulfasalazine treatment group, Sulfasalazine combined with Ulinastatin treatment group, and normal control group were arranged to drink distilled water. Seven days after treatment, relevant parameters of mice were measured: disease activity index (DAI), changes of colon histopathology, expressions of ALB, CRP and TNF-α in serum, the expressions of SOD and NF-KB in colonic mucosa. Results Comparison of activity index, CRP, TNF-α and NF-κB of mice in each group after treatment: there were the normal control group 〈 Sulfasalazine combined with Ulinastatin treatment group 〈 Sulfasalazine treatment group 〈 model control group ; comparison of ALB and SOD : there were the normal control group 〉 Sulfasalazine combined with Ulinastatin treatment group 〉 Sulfasalazine treatment group 〉 model control group. Conclusion Ulinas- tatin can significantly inhibit the expressions of NF-κB, and the therapeutic effect of UC is distinctly Superior to the use of traditional drug Sulfasalazine.
