RP-HPLC法测定香青兰有效部位中7种成分的含量 被引量:2

摘要 目的:建立RP-HPLC法测定香青兰有效部位中田蓟苷、金合欢素、木犀草素-7-O-β-葡萄糖醛酸苷等7种成分含量的方法。方法:Sunfire-C18(250 mm×4.6 mm,5μm)色谱柱;以乙腈(A)-0.2%磷酸水溶液(B)为流动相,梯度洗脱(0~5min,16%A;5~45min,16%→28%A;45~65min,28%→80%A);流速1.0m L·min^(-1);柱温35℃;检测波长330 nm。结果:木犀草素-7-O-β-葡萄糖醛酸苷、芹菜素-7-O-β-葡萄糖醛酸苷、迷迭香酸、香叶木素-7-O-β-葡萄糖醛酸苷、田蓟苷、金合欢素-7-O-β-葡萄糖醛酸苷和金合欢素分离均良好,分别在1.20~60.00μg·m L^(-1)(r=0.9999)、0.48~24.00μg·m L^(-1)(r=0.9997)、0.88~44.00μg·m L^(-1)(r=0.9999)、0.88~44.00μg·m L^(-1)(r=0.9999)、3.84~192.00μg·m L^(-1)(r=0.9999)、0.72~36.00μg·m L^(-1)(r=0.9999)和0.72~36.00μg·m L^(-1)(r=0.9997)范围内呈良好线性关系;平均回收率分别为99.29%,97.04%,99.56%,98.29%,101.18%,99.34%和95.50%,RSD值分别为0.52%,1.13%,0.55%,0.42%,0.58%,0.97%和0.60%。结论:该色谱方法灵敏、稳定、简单、准确,可用于香青兰有效部位及制剂的质量控制。 Objective: To establish a RP-HPLC method for simultaneous determination of tilianin,Acacetin,Luteolin-7-O-β-glucuronide et.chemical composition in the effective part of Dracocephalum Moldavica L.Methods:The column was Sunfire-C18(250mm×4.6mm,5μm),mobile phase was acetonitrile(A)-0.2%phosphoric acid solution(B) with linear gradient elution(0-5min,16%A;5-45min,16%→28%A;45-65min,28%→80%A);flow rate was 1.05 m L·min^-1;column temperature was 35℃ and detection wavelength was 330 nm.Results:This mathod had good linear relationship in range of 1.20-60.00μg·m L^-1(r=0.9999) for Luteolin-7-O-β-glucuronide,0.48-24.00μg·m L^-1(r=0.9997) for Apigenin-7-O-β-glucuronide,0.88-44.00μg·m L^-1(r=0.9999) for Rosmarinic acid,0.88-44.00μg·m L^-1(r=0.9999) for Diosmetin-7-O-β-glucuronide,3.84-192.00μg·m L^-1(r=0.9999) for Tilianin,0.72-36.00μg·m L^-1(r=0.9999) for Acacetin-7-O-β-glucuronide and 0.72-36.00μg·m L^-1(r=0.9997) for Acacetin.Average recoveries of Luteolin-7-O-β-glucuronide,Apigenin-7-O-β-glucuronide,Rosmarinic acid,Diosmetin-7-O-β-glucuronide,Tilianin,Acacetin-7-O-β-glucuronide and Acacetin were 99.29%,97.04%,99.56%,98.29%,101.18%,99.34% and 95.50%,respectively.RSD were 0.52%,1.13%,0.55%,0.42%,0.58%,0.97% and 0.60%,respectively.Conclusion:The RP-HPLC method was stable,simple and accurate.It can be used to control the quality of preparation and the effective part of.Dracocephalum Moldavica L..
