摘要
目的:研究乳腺癌MCF-7细胞株中PKGⅡ对抑癌基因PTEN、p21、p53、BRCA1以及BRCA2蛋白表达的影响。方法:用编码PKGⅡc DNA的腺病毒载体感染MCF-7细胞,使其高表达PKGⅡ并以相应激活剂8-p CPT-cGMP激活;蛋白质印迹法检测MCF-7细胞p53、p21、PTEN、BRCA1以及BRCA2因子表达量。结果:PKGⅡ表达增高并激活后可使乳腺癌MCF-7细胞株中抑癌基因的蛋白表达降低。结论:PKGⅡ通过降低癌细胞的活动水平从而降低抑癌基因蛋白的表达。
Objective: To investigate the effects of PKGⅡ on tumor suppressor genes including PTEN, p21, p53 ,breast cancer type 1 susceptibility protein (BRCA1) and BRCA2 in breast cancer cell line MCF- 7. Methods : MCF-7 cells were transfected with adenoviral vector encoding cDNA of PKG Ⅱ and the kina- ses were activated by respective activator. Western blotting was applied to detect the influence of PKG Ⅱ on the expression of tumor suppressor proteins, including p53, p21, FFEN, BRCA1 and BRCA2. Results: When PKGⅡ expression and activity were increased, the expression of PTEN, p21, p53, BRCA1 and BRCA2 was decreased. Conclusion: PKG Ⅱ could inhibit the expression of PTEN, p21, p53, BRCA1 and BRCA2.
