高强度聚焦超声联合N_2O对SKVO3细胞影响的实验研究 被引量:3

摘要 目的:通过比较高强度聚焦超声(high intensity focused ultrasound,HIFU)和N_2O单独或联合使用对SKVO3细胞的影响,研究HIFU联合N_2O对SKVO3的作用。方法 :将培养的SKOV3细胞分为4组并作以下处理:空白对照组:不作任何处理;N_2O组:鼓泡法通入N_2O(60 min);HIFU组:使用HIFU(250W,5 s)辐照;N_2O联合HIFU组:鼓泡法通入N_2O(60 min)并使用HIFU(250W,5s)辐照;处理后,用膜联蛋白Annexin V-FITC/PI流式方法检测细胞的死亡凋亡情况,用western blot检测Cytochrome C、Capase 3、Bax、Bcl-2和GAPDH表达情况。结果 :N_2O-HIFU组的细胞死亡和凋亡情况都显著高于其他任意组(P<0.05)。Western blot检测结果 :N_2O-HIFU组Cytochrome C、Caspase 3和Bax显著增加,Bcl-2显著被抑制。结论 :HIFU联合N_2O相较于二者单独使用能对SKVO3细胞产生更强的杀伤作用。 Objective To explore the effect of N20 with High Intensity Focused Ultrasound (HIFU) on SKVO3 through observing the influence of HIFU and N20, separated and combined, on SKVO3. Methods The cultured SKVO3 ceils were divided into 4 group: control group: without any treatment; N20 group: blowcd by N20 60 minutes; HIFU group: irradiated by HIFU (250W, 5s) and N20-HIFU group blowed by N20 60 minutes and irradiated by HIFU (250W, 5s). Flow cytometric method and Annexin V-FITC/PI binding assay were applied to detect cell death and apoptosis, and western blot test to detect Cytochrome C, Capase 3, Bax, Bcl-2 and GAPDH. Results The cell death and apoptosis in N20-HIFU group were significant higher than those in other groups(P 〈 0.05). The results of western blot showed that in N20-HIFU group, CytochromeC, Caspasc 3 and Bax increased remarkably, but Bcl-2 significantly suppressed. Conclusion HIFU combininh with N20 could enhance the killing of SKVO3 cell.
