摘要
目的评估ISOLA全自动平板接种仪使用Digest-EUR与SPUTASOL、胰蛋白酶痰消化液处理痰标本的液化和接种效果,以及不同试剂对细菌生长的影响,为选择痰消化液提供依据。方法收集2014年10月-2015年1月医院住院患者120份中等粘稠痰标本和特粘稠痰标本,同时采用三种试剂对标本进行预消化处理,评估接种效果,三种试剂分别与肺炎链球菌、流感嗜血菌菌液接触0、15、30、60、90、120min,观察不同作用时间对肺炎链球菌和流感嗜血菌生长的影响,并使用SPSS统计软件进行统计分析。结果三种试剂对两种粘度痰标本消化处理后,进行全自动标本接种均未出现吸头堵塞,消化预处理效果差异无统计学意义;在哥伦比亚血平板、中国蓝平板和巧克力平板上的接种效果差异无统计学意义;Digest-EUR和SPUTASOL两种试剂与肺炎链球菌、流感嗜血菌菌液接触不同时间后,均对生长无明显影响,胰蛋白酶作用15min后,即对肺炎链球菌、流感嗜血菌的生长产生抑制。结论三种试剂均可用于ISOLA全自动平板接种仪对痰标本的消化预处理,其中Digest-EUR和SPUTASOL消化液对肺炎链球菌和流感嗜血菌生长无明显影响,胰蛋白酶液化15min后对肺炎链球菌和流感嗜血菌的生长有明显的抑制作用。
OBJECTIVE To evaluate the effect of liquefaction, inoculation and the growth of bacteria of Digest-EUR (BioMerieux), SPUTASOL(OXOID) and trypsin (Sigma) sputum digestive reagents with the Previ Isola auto- matic flat inoculation system (ISOLA), providing a basis for the selection of sputum digestion reagents. METHODS 120 of moderate viscous sputum specimens and severe viscous sputum samples were collected in PLA General Hos- pital inpatients, from October 2014 to January 2015 , which were digested by three above reagents, to assess the efficacy of inoculation . Three reagents respectively processed Streptococcus pneumonia , Haemophilus influ- enzae bacteria at 0rain, 15min, 30min, 60rain, 90min, 120min. After that, the growth of bacteria was observed. SPSS statistical software statistical analysis was performed. RESULTS After three reagents for both of viscous spu- tum samples digestion process, the ISOLA tips were not clogged. The digestion pretreatment effect was no signifi- cantly different. There was no significant difference effect on three kinds of Colombia blood plates, Chinese blue plates and chocolate plates . Digest-EUR and SPUTASOL had no effect on the growth of S. pneumoniae, H. in- fluenzae bacteria at different times, while trypsin suppressed S. pneumoniae and H. influenzae growth at 15 min. CONCLUSION The three reagents are available for pre-digested sputum specimens with ISOLA. Digest-EUR and sPUTASOL have no significant effect on the growth of S. pneumoniae and H. influenzae. Howere, trypsin liquefaction after 15min inhibits S. pneumoniae and H. influenzae growth.
