摘要 磷脂肌醇(PI3K)-蛋白激酶B(PKB,Akt)-雷帕霉素靶体蛋白(mTOR)通路在细胞的存活、增殖和分化等过程中发挥关键作用。PI3K-Akt-mTOR信号通路的异常激活和人类肿瘤的发生密切相关,许多靶向该通路的小分子抑制剂已经进入临床试验中。本文将对近年来PI3K-Akt-mTOR信号通路与肿瘤的关系及PI3K-Akt-mTOR抑制剂抗肿瘤作用研究进展,并结合本课题组的相关研究成果进行综述。 Aberrant activation of the phosphatidylinositol 3-kinase( PI3K)-protein kinase B( PKB,Akt)-mammalian target of rapamycin( m TOR)pathway is commonly observed in human cancer and is critical for cell survival,proliferation and differentiation. A variety of small molecule inhibitors targeting PI3K-Akt-m TOR pathway are under clinical studies. This review will summarize the recent studies in terms of the PI3K-Akt-m TOR signaling pathway and cancer,research progress of the antitumor activity possessed by PI3K-Akt-mTOR inhibitors,as well as the recent research in the related field conducted by our group.
