周期素依赖性激酶抑制因子3在11种常见肿瘤中的蛋白表达模式

摘要 目的 观察周期素依赖性激酶抑制因子3(CDKN3)基因在不同肿瘤的表达模式,探讨其在不同肿瘤发生发展中的角色.方法 应用免疫组织化学技术对11种常见肿瘤类型进行CDKN3蛋白表达及组织定位进行检测.结果 发现CDKN3蛋白定位呈现胞浆阳性,少量核阳性,在各类肿瘤无明显差异.直肠癌(4.40±0.26)、结肠癌(4.10±0.55)及胰腺癌(4.00±0.50)细胞中表达丰度高;而在肺癌(3.30±0.35)、食道癌(3.60±0.30)、胃癌(3.20±0.50)、卵巢癌(3.30±0.40)及肝癌(3.00±0.55)中呈现中等表达,在胆管癌(2.30 ±0.42)、乳腺癌(2.20 ±0.36)及肾癌(2.00 ±0.15)中表达较弱.结论 CDKN3在各类肿瘤中表达强弱有明显差异,这说明其在肿瘤发生发展中的作用可能因肿瘤背景而异. Objective To elucidate the expression pattern of Cyclin dependent kinase inhibitor 3(CDKN3) gene in different tumors and reveal its role in the progression of different tumors.Methods The immunohistochemical techniques were used to detect CDKN3 protein expression in 11 kinds of tumor.Results CDKN3 protein is dominative expression in cytoplasm and a little positive in cell nucleus.There are no significant differences of CDKN3 cell location among these tumors,rectal cancer (4.40 ± 0.26),Colon cancer (4.10 ±0.55) and pancreatic cancer (4.00 ±0.50) cells highest express CDKN3 protein;lung cancer (3.30 ± 0.35),esophagus cancer (3.60 ± 0.30),gastric cancer (3.20 ± 0.50) ovarian cancer (3.30 ± 0.40) and liver cancer (3.00 ± 0.55) have moderate CDKN3 expression;Breast cancer (2.20 ±0.36),cholangiocarcinoma (2.30 ±0.42) and kidney cancer (2.00 ±0.15) have lowest expression of CDKN3.Conclusion Expression pattern of CDKN3 in various types of tumors was significantly different,which maybe explain the different role of CDKN3 in different cancers.
