摘要
为探讨Desmin在结直肠癌组织中的表达情况及其临床意义,本研究使用组织芯片、免疫组织化学及Westernblot等方法检测结直肠癌组织及癌旁正常黏膜组织中的Desmin表达情况,分析Desmin表达与结直肠癌临床病理参数及预后的相关性。结果显示,Desmin在结直肠癌组织和癌旁正常黏膜组织中的表达水平存在明显差异,其在结直肠癌组织中表达水平明显升高。Chi—square分析结果显示,Desmin的表达与T分期(P〈0.05)、N分期(P〈0.05)、M分期(P〈O.05)、TNM分期(P〈0.05)、分化程度(P〈0.05)以及术前血CEA水平(P〈0.05)呈正相关,与性别、年龄、肿瘤位置以及术前血CA19—9水平无明显相关性(P〉0.05)。Knplan—Meier生存分析显示,与Desmin高表达患者相比,Desmin低表达患者总的3年生存率(3-yearOS)明显增高(Log—rank检验,P〈0.001)。结果表明,Desmin与结直肠癌的发生发展密切相关,并可能成为一种潜在性的肿瘤标志物。
This study was to explore the expression of Desmin in colorectal cancer tissue and its clinical significance;detected the expression of Desmin in colorectal cancer tissue and adjacent cancer normal mucosa tissue by the method of tissue microarray, immunohistochemistry, and Western blot; analyzed the relationship between the Desmin expression and clinicopathologieal parameters & prognosis of colorectal cancer. Results show that there was obvious difference on the Desmin expression in colorectal cancer tissue and adjacent cancer normal mucosa tissue ( P 〈0.05) and the expression in colorectal cancer tissue was significantly higher. Chi-square analysis results show that there was positive correlation between Desmin expression and T stage ( P 〈0.05), N stage ( P 〈0.05), M stage ( P 〈0.05),TNM stage ( P 〈0.05),differentiation degree ( P 〈0.05) & preoperative serum CEA level ( P 〈0.05); but no significant correlation between Desmin expression and sex, age, tumor location & preoperative serum CA19-9 (P 〉 0.05). Kaplan-Meier survival analysis show that compared with Desmin high expression patients,the total 3-year survival rate (3-year OS) of Desmin low expression patients was significantly higher ( Log-rank detection, P〈0.001).In conclusion,there is close relationship between Desmin and the occurrence & development of colorectal cancer and Desmin probably become a potential tumor marker.
