头针对脑缺血再灌注大鼠脑微血管层粘连蛋白影响的动态研究 被引量:1

摘要 目的:动态观察头针对脑缺血再灌注大鼠脑微血管层粘连蛋白的影响,以探讨头针抗脑缺血再灌注损伤的可能作用机制。方法:130只SD大鼠,随机分为假手术组、模型组和针刺组。采用改良的Zea Longa线栓法制作脑缺血再灌注模型,针刺组以头穴透刺的方法,选取百会(Du20)透曲鬓(GB7)针刺治疗30 min,每12 h针刺1次。应用免疫组化和免疫印迹检测方法,比较3组大鼠不同时间点脑微血管层粘连蛋白阳性表达与含量的变化。结果:模型组大鼠各时间点脑微血管层粘连蛋白表达较假手术组明显降低;针刺组在再灌注4、12 h,梗死灶周围区的层粘连蛋白阳性表达及含量均与模型组相似(P>0.05);再灌注24、48、72 h较模型组增加(P<0.05),但仍低于假手术组(P<0.05)。结论:早期头针治疗能增加缺血侧脑微血管层粘连蛋白的表达,并随着针刺治疗时间的延长,其效果明显提高。通过上调层粘连蛋白保护血脑屏障,可能是头针拮抗脑缺血再灌注损伤的作用途径之一。 Objective: To investigate the effect of scalp acupuncture on microvascular Laminin expression in the rats with cerebral ischemia reperfusion for exploring the interaction mechanisms. Methods: 130 Male SD rats were randomly divided into three groups: sham operation group,model group and scalp acupuncture group. Cere-bral ischemia / reperfusion model was induced by modified Longa's method. Scalp acupuncturing group was treated with Baihui- Qubin( Du20- GB7) acupuncture for 30min,once per 12 h. The positive expression and protein content of Laminin were determined by immunohistochemistry and Western blot Methods. Results: Laminin expression and content in model group rats' brain at each time- point were lower to compare with sham group,the expression and content of Laminin about infarction in the scalp acupuncture group were similar with the model group in IR4 h and IR12 h( P〈0. 05),but which were higher obviously than those of model group in IR24 h,IR48 h and IR72 h( P〈0. 05),and in the same time-point,the expression and content of Laminin were still lower than those in the sham group( P〈0. 05). Conclusion: Scalp- acupuncture therapy can promote the expression of Laminin in rats with acute cerebral ischemia /reperfusion,and that was further increased with the treatment time prolonging. Scalp- acupuncture therapy can ameliorate cerebral ischemia reperfusion injury by possibly enhancing Laminin expression and protecting blood-brain barrier.
