IFN-γ参与右美托咪定在单关节炎大鼠模型中的镇痛作用 被引量:3

Spinal IFN-γ Involves in Dexmedetomidine-induced Analgesia in Monoarthiritis Rats

摘要 目的:探讨IFN-γ是否参与了右美托咪定在大鼠单关节炎模型中的镇痛作用。方法:选用完全弗氏佐剂(CFA)诱导的大鼠单关节炎模型,应用Von Frey纤毛检测大鼠造模以后以及造模并且鞘内连续给予右美托咪定后大鼠后足机械痛敏的变化。应用免疫荧光组织化学实验检测大鼠脊髓背角IFN-γ的定位;应用蛋白质免疫印迹检测大鼠脊髓背角IFN-γ表达的变化。结果:大鼠单关节炎模型建立后,脊髓背角IFN-γ的表达显著上调;大鼠脊髓IFN-γ定位于脊髓背角浅层;鞘内连续给予右美托咪定抑制了单关节炎大鼠的机械痛敏,并且减少了单关节炎大鼠脊髓背角IFN-γ的表达。结论:脊髓背角IFN-γ的表达水平与右美托咪定对单关节炎大鼠的镇痛作用有关。 Objective: To investigate whether the analgesia effect of dexmedetomidine is associated with spinal IFN-3, in Monoarthiritis rats' model. Methods: The rat model of complete Freund's adjuvant (CFA)-induced ankle joint monoarthritis (MA) was established. Von Frey hair was used to test the change of mechanical allodynia after CFA injection or CFA injection and dexmedetomidine lumbar puncture (LP) injection. Immunohistochemistry and western blot was used to test the location and the expression oflFN-y in the spinal dorsal horn. Results: Immunohistochemistry and western blot showed the increased IFN-γ expression in the spinal dorsal horn. Immunofluorescence revealed that IFN-γ was localized to the superficial lamina of the spinal dorsal horn. Inthrathecal administration of Dexmedetomidine inhibited the pain behavior and reduced WN-γ up-regulation in the ipsilateral spinal dorsal horn. Conclusions: The spinal IFN-γ is involved in the anti-nociceptive effect of dexmedetomidine in monoarthritis rats.
