摘要
目的:探讨HPV基因分型对HPV E6/E7 mRNA表达的影响。方法:回顾分析227例接受HPV E6/E7 mRNA检测的HPV病毒感染患者,高危型HPV病毒感染者204例,低危型HPV病毒感染者23例。PCR导流杂交方法检测HPV基因分型,支链DNA法检测HPV E6/E7 mRNA载量。结果:高危型HPV感染患者宫颈上皮细胞内HPV E6/E7 mRNA表达载量[201.15(0.00,671.98)]较低危型HPV感染患者[0.00(0.00,16.00)]增高(P<0.05)。多因素回归分析发现HPV52(β=6936.219)、HPV18(β=4551.115)、HPV45(β=5178.912)三个高危亚型可影响宫颈上皮细胞内HPV E6/E7 mRNA载量(P<0.05)。结论:高危型HPV52、HPV18、HPV45病毒感染可影响宫颈上皮细胞内HPV E6/E7 mRNA的表达,进而促进宫颈恶性病变的发生和发展。
Objective: To investigate the association of human papillomavirus( HPV) genotyping with HPV E6 /E7 mRNA expression.Methods: Retrospective analysis was performed in 227 HPV infection cases received HPV E6 / E7 mRNA detection,including 204 cases of high-risk HPV types and 23low-risk HPV types.HPV genotype was detected by PCR diversion hybridization,and HPV E6 / E7 mRNA loading were determined by branched DNA assay.Results: The HPV E6 / E7 mRNA loading in cervical epithelial cells was significantly increased in patients infected with high-risk HPV types [201.15( 0.00,671.98) ]compared to those infected with low-risk HPV types [0.00( 0.00,16.00),P〈0.05].Multiple regression analysis indicated that the HPV E6 / E7 mRNA loading was affected by HPV52( β = 6936.219),HPV18( β = 4551.115) and HPV45( β = 5178.912)( P〈 0.05). Conclusion: Infection with the high-risk types of HPV52,HPV18 and HPV45 may lead to HPV E6 / E7 mRNA loading in cervical epithelial cells,and further result in the development of cervical malignancies.
