类风湿关节炎患者外周血miR-146a及miR-155的表达及临床意义 被引量:8

摘要 目的:探讨类风湿关节炎患者外周血淋巴细胞中miR-146a及miR-155的表达及临床意义。方法:应用实时荧光定量PCR法检测30例类风湿关节炎患者及30例健康者外周血淋巴细胞中miR-146a、miR-155的表达水平,并且与类风湿关节炎患者红细胞沉降率、C-反应蛋白、DAS28评分、免疫球蛋白M型类风湿因子进行相关性分析。结果:miR-146a及miR-155在类风湿关节炎患者外周血淋巴细胞中的相对表达水平均高于对照组,且差异有统计学意义(P<0.05)。类风湿关节炎患者miR-146a、miR-155的表达水平与红细胞沉降率、C-反应蛋白、DAS28呈正相关(P<0.05),与免疫球蛋白M型类风湿因子无相关性(P>0.05)。结论:类风湿关节炎患者外周血miR-146a和miR-155表达上调,其表达水平与类风湿关节炎病情活动相关,提示miR-146a和miR-155可作为类风湿关节炎疾病活动的有效指标。 Objective:To investigate the expression and clinical significance of miR-146a and miR-155 in peripheral blood lymphocytes of patients with rheumatoid arthritis.Methods:The real time fluorescent quantitative PCR was used to detect the expression levels of miR-146a and miR-155 in peripheral blood lymphocytes of 30 cases of rheumatoid arthritis and 30 healthy people.A correlation analysis was made between above detected levels and patients' erythrocyte sedimentation rate,C-reactive protein,DAS28 score and immunoglobulin M rheumatoid factor.Results:The relative expression levels of miR-146a and miR-155 in the peripheral blood lymphocytes of patients with rheumatoid arthritis were higher than those of the control group,and the difference was statistically significant(P〈 0.05).The expression levels of miR-146a and miR-155 were positively correlated(P〈 0.05)with erythrocyte sedimentation rate,C-reactive protein and DAS28 and had no correlation(P〉 0.05)with immunoglobulin M rheumatoid factor.Conclusion:The expression of miR-146a and miR-155 in peripheral blood of patients with rheumatoid arthritis up-regulates and the expression level is related to disease activity,suggesting that miR-146a and miR-155 can be used as effective indicators of disease activity of rheumatoid arthritis.
