不同电针刺激强度对急性脑缺血再灌注大鼠NSS与脑组织含水量的影响 被引量:5

The influence of different intensity of electroacupuncture on NSS and the content of tissue water in cerebral ischemia reperfusion rats

摘要 目的研究不同电针刺激强度对急性脑缺血再灌注大鼠神经功能缺损评分(NSS)与脑组织含水量的影响。方法将50只8周龄健康大鼠随机分为5组:正常组、假手术组、模型对照组、弱电针刺激组、强电针刺激组,每组10只,后3组建立大脑中动脉缺血(MCAO)再灌注模型,治疗组在缺血再灌注(I/R)6 h后开始针刺。24 h二次针刺后行神经功能缺损评分,然后断头取脑,采用干湿法测定脑组织含水量;对各指标进行统计学分析。结果(1)各干预组大鼠神经功能缺损情况均较前改善(P<0.01),但各组间无明显统计学差异(P>0.05)。(2)弱电针刺激组脑组织含水量较模型对照组无明显变化(P>0.05),强电针刺激组脑组织含水量较模型对照组明显降低(P<0.05),强电针刺激组与弱电针刺激组脑组织含水量无明显差异(P>0.05)。结论急性脑缺血再灌注大鼠神经功能缺损评分在24 h会有所改善,因此不能说明24 h大鼠NSS的改善是电针的作用效果。强电针刺激能通过降低MCAO再灌注模型大鼠脑组织含水量从而对急性脑缺血再灌注大鼠起保护作用。 Objective To study the changes of neurological deficit score( NSS) and the content of tissue water through the different intensity of electroacupuncture( EA) in rats with acute cerebral ischemia reperfusion. Methods 50( fifty) eight-week-old and health rats were equally divided into 5 groups randomly: the normal group,the sham-operation group,control group,weak EA stimulation group and strong EA stimulation group. The middle cerebral artery occlusion( MCAO) and reperfusion model were established in the latter three groups. After 6 hours,the weak electroacupuncture stimulation and strong electroacupuncture stimulation were implemented in two model groups,respectively,which were weak EA stimulation group and strong EA stimulation group,while no treatmentment was on the other model group as control group. After 24 hours,the same acupunctures were implemented in those two treament groups,then the neurological deficit were scored and the brains were collected. The brain water content were detected by wet and dry method and each indicator were analysised. Results( 1) The NSS were sig-nificantly improved in each interventional group( P〈 0. 01) compare with the treatment,however,there were no differences in each group( P〉 0. 05).( 2) Compared with the control group,there were no change in the content of tissue water in brain of the weak EA group( P〉 0. 05). However,it decreased significantly( P 〈0. 05) in the strong EA group. Compared with the weak EA Group,there were no obvious differences in the content of tissue water in brain of the strong EA group( P〉 0. 05). Conclusion The neurological deficit score of acute cerebral ischemia-reperfusion rats was improved in 24 hours,so it can not be explained that the improvement of NSS in 24 h is the effect of electroacupuncture. Strong electro-acupuncture stimulation can protect rats from cerebral ischemiareperfusion injury by decreasing the water content of brain tissue in MCAO-reperfusion model rats.
