IBDV感染雏鸡法氏囊和脾脏中抗病毒基因Mx的表达变化 被引量:2

摘要 Mx蛋白是由干扰素诱导产生的在多种脊椎动物中发挥抗病毒作用的效应分子。为探究雏鸡感染传染性法氏囊病病毒(IBDV)后鸡抗病毒基因Mx m RNA在法氏囊和脾脏中的表达变化,采用IBDV感染雏鸡70只,分别于感染后0、24、72、120、168、192 h随机选取6只,收集法氏囊和脾脏,利用q RT-PCR技术检测IBDV感染后不同时间点鸡Mx m RNA表达变化。结果显示,雏鸡感染IBDV后,法氏囊和脾脏中Mx m RNA表达水平均极显著高于对照组(P<0.01),对照组各时间点之间几乎无变化;且随着感染时间延长,法氏囊和脾脏中Mx m RNA表达量呈先增高后降低的趋势,法氏囊和脾脏分别在72和24 h Mx表达量达到最高,是对照组的66.13和25.40倍。结果表明,法氏囊和脾脏中Mx的转录水平随着IBDV在法氏囊中的增殖而迅速升高,而IBDV减少后Mx的转录水平下降;脾脏中Mx对IBDV感染的转录水平变化比法氏囊更为快速。 Mx Protein is a vital antiviral effector induced by type Ⅰ interferon in many vertebrates. To explore the expression change of Mx m RNA in Leghorn chicken during infectious bursal disease virus(IBDV)infection,seventy Leghorn chickens were infected with the standard strains of IBDV preserved in our laboratory. Bursa of Fabricius and spleen of 6 chickens randomly selected were collected at 0,24,72,120,168,192 h after infected with IBDV,respectively.Real- time quantitative PCR was applied to detect the expression levels of Mx m RNA from bursa of Fabricius and spleen of chicken in the IBDV infection group and the control group. The results showed that the expression levels of Mx m RNA in the infected groups were significantly higher than these in bursal of Fabricius and spleen of control group(P〈0.01),and there were almost no changes in control group at different time points. The expression patterns of Mx m RNA in bursa of Fabricius and spleen of chicken had a trend of increasing at first and then decreasing during IBDV infection,and Mx m RNA expression levels reached the highest at 72 h in bursa of Fabricius and at 24 h in spleen,which were66.13 and 25.40 fold higher than that of control group,respectively. These results indicated that the expression levelsof Mx m RNA in bursa of Fabricius and spleen were rapidly increased by following IBDV replication,and their levels decreased with reduced IBDV. And the expression patterns of Mx m RNA in bursa of Fabricius and spleen existed a little in difference. The transcriptional changes of Mx in spleen were more rapid than that in bursa of Fabricius.
