檀香叶中牡荆苷与异牡荆苷的提取纯化工艺优选

Optimization of Technical Processes for Extraction and Purification of Vitexin and Isovitexin from Leaves of Santalum album

摘要 目的优选檀香叶中牡荆苷与异牡荆苷的提取和纯化工艺。方法以牡荆苷与异牡荆苷的提取率为指标,通过正交试验考察料液比、提取时间、乙醇体积分数、提取次数等因素的影响,优选提取工艺。通过单因素试验考察影响大孔吸附树脂的吸附率、解析率的多种因素,优选檀香叶中牡荆苷与异牡荆苷的纯化工艺。结果优选所得提取工艺为50℃恒温超声提取,料液比1∶8,提取时间为40 min,提取溶剂为60%(V∶V)乙醇,提取次数为3次。纯化工艺为AB-8大孔吸附树脂,上样液浓度为0.25 g/ml(生药量/溶剂);洗脱顺序为:10%乙醇除杂,洗脱体积约10~12 BV(至流出液无色),再换用30%乙醇,洗脱6 BV,洗脱速度为2 BV/h。结论提取与纯化工艺操作简便、稳定可行。 Objective To optimize technical processes for extraction and purification of vitexin and isovitexin from leaves of Santalum album. Methods Using orthogonal design,taking the extraction percentage of vitexin and isovitexin as indexes,the extraction and purification conditions of vitexin and isovitexin from leaves of Santalum album were optimized respectively. Results The optimal technical process for extraction of vitexin and isovitexin was determined to be ultrasonic extraction with 60% ethanol at 50℃ for three times,the extracting time was 40 min,the ration of raw materials to solvent was 1: 8. The optimized method for purification of vitexin and isovitexin were as follows: AB-8 macroporous absorption resin,sample concentration was 0. 25 g / ml( dry herbs to solvent),the order of eluting was 10% ethanol for 10 ~ 12 BV( until the effluent colourless) then to change to 30% ethanol for 6 BV. Conclusion The optimal technical processes for extraction and purification of vitexin and isovitexin are simple and practical.
