秦苓液对尿酸性肾病大鼠肾功能及肾组织病理改变的影响 被引量:7

Effect of Qinling Liquid on Renal Function and Pathological Changes of Renal Tissue in Rats with Uric Acid Nephropathy

摘要 目的:观察秦苓液对尿酸性肾损害大鼠肾功能及肾组织病理改变的影响,探讨其对尿酸性肾病(UN)肾脏损伤的保护作用及作用机制。方法:采用腺嘌呤灌胃伴酵母饲料喂养的方法造模,将成模大鼠随机分为正常组,模型组,别嘌呤醇组(23.33 mg·kg^(-1)·d^(-1)),秦苓液大、中、小剂量组(36.4,18.2,9.1 g·kg^(-1)·d^(-1)),连续干预8周。分别于0,2,4,6,8周检测各组大鼠24 h尿蛋白(u PRO/24 h),血尿酸(s UA),血肌酐(s CR)和血尿素氮(BUN);干预6周和8周后分别处死各组大鼠1/2量,取肾脏进行苏木素-伊红和过碘酸雪夫氏染色,光镜下观察。结果:与正常组比较,模型组大鼠u PRO/24 h,s UA,s CR,BUN明显升高,肾组织出现较明显的病理改变;与模型组比较,秦苓液各剂量组大鼠u PRO/24 h,s UA,s CR,BUN有不同程度的降低,肾组织病理损伤明显减轻。结论:秦苓液能显著改善UN模型大鼠肾功能,减轻肾组织病理损伤,保护肾脏。 Objective: To observe the effects of Qinling liquid on renal function and pathological changes of renal tissue in rats with uric acid nephropathy,and to discuss its protection mechanism for renal injury.Method: The rat model was induced by gavaging adenine and feeding yeast. Model rats were randomly divided into the normal group,the model group,the positive group( 23. 33 mg·kg^- 1·d^-1of allopurinol) and high-dose,medium-dose,low-dose groups of Qinling liquid( 36. 4,18. 2,9. 1 g·kg^- 1·d^-1),the normal group and the model group were daily gavaged with distilled water. At the timepoint of 0,2,4,6,8 weeks,the levels of serum uric acid( s UA),serum creatinine( s CR),blood urea nitrogen( BUN) and 24 h urine protein( u PRO/24h) were detected. At the end of 6thand 8thweek,half of rats in each group were sacrificed and the kidney was harvested for hematoxylin-eosin and periodic acid-schiff staining. Result: Compared with the normal group,the levels of u PRO/24 h,s UA,s CR and BUN in the model group were dramatically inceased,pathological changes of renal tissue were serious. Compared with the model group,the levels of u PRO/24 h,s UA,s CR and BUN in each group of Qinling liquid were deceased at different degrees,and the pathological changes of renal tissue were alleviated. Conclusion: In model rats of uric acid nephropathy,Qinling liquid can significantly improve renal function and relieve renal pathological damage for kidney protection.
