摘要
在pH=4.5的B-R缓冲溶液中,Fe(Ⅲ)与5-磺基水杨酸配位生成紫红色配合物,使得体系在257nm和342nm处产生2个共振散射峰。在该体系中加入肝素钠后,在静电引力作用下其与配合物结合形成离子缔合物,从而使得体系在257nm和342nm处的共振散射信号减弱。在优化实验条件下,在0.05-15.0μg/mL质量浓度范围内,体系的共振散射信号△I与肝素钠的浓度之间有较好的线性关系(相关系数r=0.9976),其检出限为39.9ng/mL。该方法可直接用于测定肝素钠注射液中肝素钠的含量,回收率为98.9%-102.2%。
In pH=4.5B-R buffer solution,Fe(Ⅲ)coordinated with 5-sulfosalicylic acid dihydrate to produce aubergine coordination compounds,generating two resonance scattering peaks at 257 nm and 342 nm respectively.Heparin sodium could combine with the complex to form ion-association complexes and weaken the resonance scattering signal.Under the optimum experimental conditions,there was a good linear relationship between the change of resonance scattering signal and the concentration of heparin sodium in the range 0.05-15.0μg/mL and the detection limit was 39.9ng/mL.This method has been directly used for the determination of heparin sodium with the recovery of 98.9%-102.2%.
