摘要 目的开发用大孔树脂从发酵液中分离纯化A-40926B_0的工艺。方法采用大孔吸附树脂吸附解吸-脱色-冻干的分离纯化方法。结果 HZ816树脂为最佳吸附剂,上柱液最佳p H为7.0,树脂吸附饱和后,先用30%乙醇洗涤,再用50%乙醇解吸,解吸收率达90.2%。所得产品纯度大于70%,收率大于60%。结论此工艺安全性高,收率稳定,适合工业化生产。 Objective To develop the separation and purification processes of A-40926B0 with macroprous resin. Methods The separation and purification processes ofA-40926B0 was studied by macroprous adsorption resin, decoloration and freeze drying successively. Results HZ816 resin was selected as the best adsorbent. The optimum pH value of fermentation broth was 7.0 before transfering to the column. After adsorption saturation, with 30% ethanol washing first, then with 50% ethanol desorption, the desorption rate ofA-40926B0 was 90.2%. The purity of the final product was over 70% and the yield of A-40926B0was above 60%. Conclusion The process is high safety, stable yield, and is suitable for industrial production.
