摘要
目的:观察黄芩苷对皮质酮诱导抑郁小鼠海马中神经营养素(Neurotrophins,NT)-3的mRNA表达水平的影响。方法:连续21d皮下注射皮质酮(40mg/kg)建立抑郁小鼠模型,采用糖水实验和强迫游泳实验评价小鼠抑郁样行为,采用实时荧光定量PCR方法检测小鼠海马内NT-3的mRNA表达水平。结果:皮质酮组小鼠糖水摄取比例显著下降,强迫游泳实验中不动时间明显延长,海马内NT-3的表达显著增高;黄芩苷逆转了皮质酮诱导的抑郁样行为,并降低海马NT-3的表达水平。结论:降低海马内NT-3的表达,可能是黄芩苷的抗抑郁机制之一。
Objective To observe the effects of baicalin on the mRNA expression of Neurotrophins (NT) -3 in the hippocampus of mice with depression induced by corticosterone.Methods Continuous subcutaneous injection of corticosterone (40 mg/kg)for 21 days to establish the mice model of depression.The sucrose preference test and forced swimming test were used to evaluate the depression -like behavior.The real -time quantitative PCR was used for detecting the mRNA expression of NT -3.Results The mice treated with corti-costerone showed the decreased sucrose preference and prolonged immobility time,as well as increased mRNA expression of NT -3 in hippocampus.Baicalin significantly reversed the depression -like behaviors induced by corticosterone,and restored the mRNA expression of NT -3 in hippocampus.Conclusion The antidepressant mechanism of baicalinmaybe related to restore the level of NT -3.
