超滤法结合HPLC法测定白藜芦醇的血浆蛋白结合率

Determination of Plasma Protein Binding Rate Of Resveratrol by Ultrafiltration and HPLC

目的：建立测定白藜芦醇与大鼠、小鼠、家兔血浆蛋白结合率的方法，并测定其结果，为白藜芦醇的药代动力学及临床用药提供参考。方法：采用超滤法结合 HPLC 法对白藜芦醇与大鼠、小鼠和家兔血浆的蛋白结合率进行测定。结果：白藜芦醇在0.521～22.97μg 的线性关系良好，标准曲线为 Y ＝11.434X －0.0409（r ＝0.9995）。白藜芦醇与大鼠血浆、小鼠血浆和家兔在0.504μg／mL、5.04μg／mL、20.16μg／mL 的含药血浆中的蛋白结合率为（90.32±0.09）％、（89.32±0.04）％、（91.52±0.12）％。结论：该方法重复性和专属性好，操作简单、快捷，能够满足样品定量分析测定要求；测定结果表明，白藜芦醇与血浆蛋白有很强的结合，且在考察范围内蛋白结合率对血药浓度无明显的依赖性。

Objective To establish a method for the determination of resveratrol of the plasma protein binding rate in rats,mice and rabbits.To provide references for the pharmacokinetics and clinical use of resveratrol.Methods The ultrafiltration was employed to determine the plasma protein binding rate of resveratrol in in rats,mice and rabbits.Results The calibration curve of resveratrol was linear within the ranges of 0.521 -22.97μg,the calibration curve was Y=11.434X -0.0409 （r =0.999）.The average plasma pro-tein binding rates of resveratrol with rats,mice and rabbits were （90.32 ±0.09）%、（89.32 ±0.04）%、（91.52 ±0.12）% in the plasma concentrations of 50.504μg/mL,5.04μg/mL,20.16μg/mL respectively.Conclusion The method has high sensitivity,good specificity and reproduction,with simple management thus fulfilling the requirement.Resveratrol shows a high binding power to plasma protein,and this was independent of the investigated concentrations and the different species.
