脂联素在结直肠癌患者中的表达及其对结直肠癌细胞增殖和Caspase-3表达的影响

Expression of adiponectin in patients with colorectal cancer and its influence on colorectal cancer cell proliferation and caspase-3 expression

目的测定结直肠癌(CRC)患者血清中脂联素水平,观察脂联素对人结直肠癌细胞LoVo增殖及凋亡相关蛋白Caspase-3的影响,初步探讨其在CRC中的作用机制。方法酶联免疫吸附试验检测CRC组及健康对照组血清脂联素水平,活细胞计数法试剂盒(cell counting kit-8,CCK-8)细胞增殖试验检测重组人脂联素对LoVo细胞增殖的影响,Western blot检测重组人脂联素作用细胞后Caspase-3蛋白的表达。结果CRC组血清脂联素水平(3.5±2.5)μg/ml低于健康对照组(4.6±2.1)μg/ml,差异有统计学意义(P<0.05)。与健康对照组比较,重组人脂联素作用LoVo细胞后,对细胞增殖有抑制作用,差异有统计学意义(P<0.05),活化的Caspase-3蛋白表达量增加,且有剂量-效应和时间-效应。结论血清脂联素水平与CRC发病风险密切相关,重组人脂联素可以抑制LoVo细胞生长,呈时间-剂量依赖性,其机制可能与Caspase-3活化相关。

Objective To detect the serum adiponectin level in patients with colorectal cancer（CRC）, and to observe the effect of adiponectin on colorectal cancer LoVo cell proliferation and apoptosis-related protein caspase-3, and preliminarily discuss the mechanism. Methods ELISA was used to detect the serum adiponectin levels of colorectal cancer group and healthy control group. CCK 8 test was used to detect the influence of recombinant human adiponectin on LoVo cell proliferation. Western blot was adopted to detect the effect of recombinant human adiponectin on caspase-3 protein expression. Results The serum adiponectin level in the CRC patients [（3.5 ± 2.5） μg/ml] was lower than that of the control group [（4.6 ± 2.1） μg/ml], the difference was statistically significant（P〈0.05）. Compared with the control group, recombinant human adiponectin significantly inhibited proliferation of LoVo cells（P〈0.05）. Adiponectin significantly increased the expression of activated caspase-3 in the cells in a dose- and time-dependent manner. Conclusions Serum adiponectin level is closely associated with colorectal cancer risk. Recombinant human adiponectin can inhibit the proliferation of LoVo cells, and the effect is time- and dose-dependent. Its mechanism may be associated with caspase-3 activation.
