贵州地方山羊RBP4基因多态性及生物信息学分析 被引量:3

Analysis on polymorphism and bioinformatics of retinol binding protein 4 gene(RBP4) in Guizhou native goat

摘要 [目的]对贵州黑山羊和黔北麻羊进行RBP4基因多态位点的筛选,为从分子水平探究RBP4基因对动物繁殖性能的调控机制提供理论依据。[方法]采用DNA混池结合直接测序法分析RBP4基因外显子4在贵州黑山羊和黔北麻羊中的单核苷酸多态性,利用生物信息学软件分析SNP位点对RBP4基因mRNA二级结构的影响。[结果]RBP4基因外显子4在贵州黑山羊和黔北麻羊中均存在4-T214C突变位点,且为同义突变。生物信息学分析结果表明,外显子4-T214C突变导致RBP4基因mRNA二级结构发生变化,自由能降低,结构稳定性增大。[结论]在贵州黑山羊和黔北麻羊中检测到RBP4基因外显子4的1个SNP位点,该突变位点影响了RBP4基因mRNA的二级结构。 [ Objective]To screen the single nucleotide polymorphism of RBP4 gene in Guizhou Black goat and Qianbei Ma goat and provide a theoretical base for exploring regulatory mechanism of RBP4 gene on animal reproductive performance from molecular level. [ Methods] DNA pool combined with direct sequencing method were used to analyzed the SNPs in the exon 4 of RBP4 gene in Guizhou Black goat and Qianbei Ma goat, bioinformatics software was applied to predict the change on the mR- NA secondary structure of RBP4 gene, which was caused by the existence of SNPs. [ Results] 4 -T214C mutation was deteced in exon 4 of RBP4 gene in both Guizhou Black goat and Qianbei Ma goat, and it belonged to sense mutation. Moreover,bioinformatics result indicated that 4 -T214C change the mRNA secondary structure of RBP4 gene,leading to lowered free energy and increased structural stability. [ Conclusion] 1 SNP locus (4 -T214C) were founded in exon 4 of RBP4 gene in both Guizhou Black goat and Qianbei Ma goat, and it affected the mRNA secondary structure of RBP4 gene.
