摘要
目的:研究紫草素对变应性鼻炎(AR)大鼠模型树突状细胞功能的免疫干预作用,探讨紫草素应用于AR的可行性。方法:健康大鼠10只处死取鼻黏膜固定为对照组;用卵清蛋白局部滴鼻法建立大鼠AR模型30只,分为观察组和模型组,模型组建模成功即脱椎处死取鼻黏膜迅速固定,观察组建模成功后予以100μg/ml紫草素治疗,12h后处死取鼻黏膜固定。用免疫组织化学技术检测3组标本中CD80、CD86、IL-10、IL-12的表达情况,组间数据采用方差检验。结果:观察组CD80、CD86分别为13.52±3.53和24.01±13.12,模型组分别为17.19±4.42和38.89±9.77;观察组IL-10、IL-12分别为6.69±1.12和44.30±9.70,模型组分别为3.61±0.23和28.80±8.20,组间差异有统计学意义(P<0.01)。结论:紫草素对AR大鼠模型中树突状细胞的异常表达可以起到纠正作用,可考虑将紫草素应用于AR治疗。
Objective:To study intervention effect of shikonin on immune function of dendritic cells using a rat allergic rhinitis model.Method:Forty rats were divided into three groups include OVA group(n=15),OVA+shikonin group(n=15),and control group(n=10).The rat allergic rhinitis model was estabilshed after OVA sensitization and local challenge.100μg/ml shikonin was spray inhalated after OVA challenge in OVA+ shikonin group.The expression of CD80,CD86,IL-10 and IL-12 was deteced by immunohistochemistry in three groups.Result:In OVA+ shikonin group,the CD80 value was 13.52±3.53,CD86 value was 24.01±13.12;in OVA group,the CD80 value was 17.19±4.42 and the CD86 value was 38.89±9.77.The IL-10 value was 6.69±1.12 and 3.61±0.23 in OVA group and OVA+ shikonin group,respectively.The IL-12 value was 44.30±9.70and28.80+8.20 in OVA group and OVA+ shikonin group,respectively.Conclusion:The shikonin has effect on immune modulation in rat model of allergic rhinitis.
