摘要
将发病猪场仔猪小肠内容物病料经胰酶处理后接种Vero细胞,经观察CPE和特异性RT-PCR确认分离得到两株PEDV野毒株,并命名"CH/GDZH01/1311"(简称"ZH01")、"CH/GDZH02/1401"(简称"ZH02")。经扩增和测定N基因,分析得知它们属于G2-1亚群流行毒株。将ZH01和ZH02及其中连续传代的ZH02-f57毒株进行3日龄仔猪的回归试验,发现它们均有100%致死率,但ZH02-f57毒株引起的发病症状相对迟缓。该研究为PED变异毒株的弱毒疫苗研究奠定了基础。
Via inoculating contents from small intestines of diarrhea piglets from infected hog farms to the Vero cell with tryp-sin,and then confirmed by the observation of CPE and specific RT-PCR,two strains of wild type PEDV were isolated and designat-ed as "CH/GDZH01/1311"( "ZH01" for short)and "CH/GDZH02/1401"( "ZH02" for short). They belong to the G2-1 subgroupbased on phylogenetic analysis of N genes. While the two isolates and ZH02-f57 were inoculated to 3-day-old piglets,a 100%mortality was observed.However clinical symptoms and deaths caused by ZH02-f57 were the latest.This study provides the founda-tion for the attenuated PEDV vaccine based on variant strains.
