摘要
为更灵敏地进行全价鸡饲料中喹乙醇(OLA)含量的快速检测,本研究将新型标记物荧光微球与喹乙醇单克隆抗体偶联,以该复合物为检测试剂制备荧光微球免疫层析试纸条,在肉眼检测的同时,将该装置与荧光微球定量侧向层析读数仪联合使用,可进行全价鸡饲料中喹乙醇含量的定量快速检测。研究结果证明,饲料中喹乙醇的定性和定量检测限分别为100μg/kg和0.51μg/kg,IC_(50)值为1.89 ng/m L,定量检测回收率为78.71%-90.83%。该试纸条特异性强、准确性高,保存期长。定量检测只需18 min左右,肉眼检测需38 min左右,与相同抗原和抗体制备的胶体金试纸条相比,该方法可大大提高灵敏度。
In order to detect the olaquindox more sensitivily,the monoclonal antibodies of olaquindox were conjugated withthe fluorescent microspheres to build a fluorescent microspheres immunoassay strip not only for the qualitative detection but alsofor the quantitative detection for olaquindox residue in complete pellet chicken feed,when the strip was coupled to a membranestrip reader. When the qualitative assay with the LOD of 100 ng/g and the quantitative assay with the LOD of 0.51 μg/kg for feed(IC_(50)=1.89 were performed,coefficient of recovery was 78.71-90.83%.The test strip was demonstrated to have high specificity,satisfying accuracy and excellent stability.The fluorescent lateral flow assay which needs only 18 min for quantitative detection and38 min for qualitative detection was confirmed to be more sensitive than colloidal gold technique developed with the same antigenand antibody.
