摘要
目的依托哈尔滨医科大学附属第四医院泌尿外科的肿瘤标本资源,建立一个规范化、信息化的肿瘤组织标本库,为泌尿系肿瘤的基础研究提供高质量的生物样本资源。方法收集笔者医院泌尿外科2013年10月~2016年1月行手术切除泌尿系统肿瘤患者的肿瘤组织、血液样本及尿液样本。所获得的手术标本立即放置在加有RNA保存液的冻存管中,然后储存在-80℃冰箱冻存。随机抽取6例-80℃冻存的肿瘤组织进行HE染色以及RNA和蛋白质的DNA提取及分析。结果2013年10月~2016年1月,泌尿系肿瘤标本库共存储189例不同组织学类型的前列腺、膀胱、肾脏、阴茎和睾丸肿瘤的标本,200例尿液和240例血浆样本。抽样检查结果显示样本分子生物学指标稳定。结论肿瘤组织样本库可提供高质量的生物样本资源及患者完整的临床信息,在一定程度上,有助于转化医学的发展,改进肿瘤诊断及治疗方法。
Objective To provide high quality biological sample resources and improve basic research of urologic neoplasms, we establish a standardized tumor tissue bank (TTB) based on the abundant clinical resources in the Fourth Hospital of Harbin Medical University. Methods The tumor tissues, which matched with tumor - adjacent tissues removed from surgery, whole blood and urine samples were collected from patient with urologic tumor and benign who were underwent operation from October 2013 to January 2016 in the hospital. The obtained surgical specimens were immediately conserved in a tube containing RNAlater, and then stored by cryopreservation ( -80℃ ). HE staining and the quality of DNA/RNA extraction were analyzed using the randomly selected 6 cases of frozen tumor tissues. Results From October 2013 to January 2016, the TTB of the Urology stored 189 samples of different histological types, including prostate, bladder, kidney, penis and testicle malignant tumors with benign mass as control; 200 samples of urine and 240 plasma samples. Sampling results showed that molecular biology indexes of the samples were stable. Conclusion TTB provides high quality of biological sample resources and patients with complete clinical information. To some extent, the establishment of TTB also promotes the development of Translational Medicine and improves the methods of diagnosis and cancer treatment.
