摘要
目的探讨脑血疏对脑出血大鼠脉络丛上皮细胞(CPECs)中食管癌相关基因4(ECRG4)表达的影响。方法将无特定病原体级SD大鼠分为脑出血组、实验组和对照组,各10只。脑出血组及实验组均用自体血注入法制备脑出血模型,对照组用等量0.9%氯化钠注射液注入尾状核,不制备脑出血模型。实验组及对照组给予等量脑血疏灌胃治疗,脑出血组给予等量0.9%氯化钠注射液灌胃。分离纯化培养各组CPECs,并应用实时定量聚合酶链反应法和蛋白质印迹法分别检测ECRG4在各组CPECs中的表达情况。结果实验组ECRG4mRNA和蛋白表达均明显高于脑出血组和对照组[(15.503±1.460)比(1.032±0.320)、(1.548±1.390),(5.268±0.750)比(1.000±0.000)、(1.348±0.120)],脑出血组ECRG4蛋白表达明显低于对照组,差异均有统计学意义(均P〈0.05)。结论ECRG4可能参与了CPECs对脑损伤的反应,并影响了CPECs的生物学特性,早期使用脑血疏可调控ECRG4在CPECs中的表达,为CPECs在神经保护中的作用研究提供了理论依据。
Objective To analyze the effect of Naoxueshu on the expression of esopageal cancer related gene 4 ( ECRG4 ) in choroid plexus epithelial cells ( CPECs ) in intracerebral hemorrhage ( ICH ) rats. Methods Ten rats were included in ICH group, observation group and control group respectively. The ICH group and observation group were ICH models; the control group was injected 0. 9% sodium chloride in caudate nucleus. Observation group and control group were treated with intragastric administration with Naoxueshu; ICH group was treated with intragastric administration with equivalent 0. 9% sodium chloride. The expression of ECRG4 in CPECs was measured by real-time polymerase chain reaction(RT-PCR) and Western blot method. Results Expressions of ECRG4 mRNA and protein in observation group were significantly higher than those in ICH group and control group[ (15. 503 ±1. 460) vs (1.032±0.320),(1.548 ±1.390);(5.268 ±0.750) vs (1.000±0.000), (1. 348 ± 0. 120 ) ] ; the expression of ECRG4 protein in ICH group was significantly lower than that in control group ( all P 〈 0. 05 ). Conclusions ECRG4 may be related to the reaction of CPECs in cerebral injury and influ- ence biologic characteristics of CPECs. Naoxueshu can regulate the expression of ECRG4 in CPECs, providing an evidence for the neuroprotective effect of CPECs.
