摘要
目的:观察vaspin对糖尿病肾病(DN)大鼠肾组织核因子-κB(NF-κB)和转化生长因子-β_1(TGF-β_1)表达的影响。方法:将40只雄性SD大鼠随机分为对照组(10只)和造模组(30只),再将30只造模成功的DN大鼠随机分为3组:DN组、vaspin组、氯沙坦组,每组10只。干预12周后检测大鼠血肌酐(Scr)、尿素氮(BUN)和24 h尿蛋白定量。应用RT-PCR和Western blot分别检测大鼠肾组织NF-κB和TGF-β_1的基因和蛋白表达。结果:DN组大鼠的Scr、BUN和24 h尿蛋白定量均高于对照组(P<0.05),肾组织中NF-κB和TGF-β_1表达水平明显高于对照组(P<0.05);vaspin组大鼠的Scr、BUN和24 h尿蛋白定量均低于DN组(P<0.05),肾组织中NF-κB和TGF-β_1的表达水平明显低于DN组(P<0.05)。结论:Vaspin可能通过下调DN大鼠肾组织NF-κB和TGF-β_1的表达,延缓DN的进展。
Objective :To investigate the effects of vaspin on expression of NF - κB and TGF - β1 in renal tissue of diabetic nephropathy rats. Methods:40 male SD rats were randomly divided into control group, diabetic nephropathy group, losartan group, vaspin group, 10 in each group. The levels of serum creatinine, urea nitrogen and 24 - hour urine protein excretion were determined by biochemistry. The expression of NF - κB and TGF - β1 in renal tissue were determined by Real - Time PCR and Western blotting. Results:Compared with control group, the expression of NF - κB and TGF - β1 and the levels of serum creatinine, urea nitrogen and 24 - hour urine protein excretion were significantly elevated in diabetic nephropathy group ( P 〈 0.05 ). Vaspin could significantly de- crease the expression of NF - κB and TGF - β1 and the levels of serum creatinine, urea nitrogen and 24 - hour urine protein excretion as compared with diabetic nephropathy group ( P 〈 0.05 ). Conclusion: Vaspin may reduce the expression of NF - κB and TGF - β1 in renal tissue of diabetic nephropathy rats, delaying the progress of diabetic nephropathy.
