摘要
糖尿病肾病(DKD)患者常伴有脂代谢紊乱。高糖诱导的过量甘油二脂(DAG)可激活蛋白激酶C(PKC),进而激活下游通路如血管内皮生长因子(VEGF)、转化生长因子β1(TGFβ1)、纤溶酶原激活物抑制物(PAI-1)、核转录因子(NF-κB)等,加速DKD的进展。同时,研究表明神经酰胺在DKD发病过程中起重要作用。而降脂药物在糖尿病肾脏疾病的效应引起了广泛关注,其保护机制可能与阻断PKC信号通路、减少脂质中间代谢产物神经酰胺生成有关。本文通过介绍脂代谢紊乱对DKD的影响以及降脂药物保护肾脏的潜在机制,阐述脂质调控与DKD之间的密切关联。
Patients with diabetic kidney disease are often associated with lipid metabolism disorders.The hyperglycemiainduced overproduction of diacylglycerol(DAG)is one of the causes for the activation of protein kinase C(PKC),which is responsible for the activation of pathways,including the production of VEGF,TGFβ1,PAI-1,and NF-κB signaling,accelerating the development of DKD.Additionally,current studies on the role of ceramide are one of the major fields of study in DKD.And lipid lowering drugs in the clinical application of diabetic kidney disease caused widespread concern,its protection mechanism may be related to blocking the activation of PKC,reduce lipid metabolites among ceramide generated.In this paper,through the introduction of the effect of lipid metabolism disorder of DKD and the possible mechanisms of lipid-lowering drugs to protect the kidney,expounds the link between the lipid regulation and diabetic kidney disease.
