摘要 目的:观察慢性紫外线(UV)损伤对小鼠角质形成细胞(KC)角蛋白(CK)1和CK10表达的影响。方法:将实验小鼠随机分为空白对照组(20只)和UV损伤组(20只),采用UV光疗仪模拟日光中的长波紫外线(UVA)和中波紫外线(UVB)照射实验小鼠。照射从最小红斑量起始,UVB为0.07 J/cm^2,UVA为0.7 J/cm^2;每日1次,每周5 d,每周增加0.5个红斑量,连续照射9周,UVB累计照射剂量达9.45 J/cm^2,UVA累计照射剂量达94.5 J/cm^2。通过苏木精-伊红(HE)染色和间苯二酚-碱性品红染色(Weigert染色法)确定皮肤损伤,通过免疫组化方法分别对照射前(W0)、后(W9)CK1和CK10的表达进行检测。应用配对t检验和独立样本t检验对相关数据进行统计分析。结果:空白对照组小鼠W0及W9时,CK1和CK10的表达差异均无统计学意义(P均>0.05)。UV损伤组小鼠W0和W9时,CK1和CK10的表达差异均有统计学意义(P均<0.05)。且UV损伤组小鼠CK1和CK10的表达呈明显的定位改变。结论:多次UV照射能导致小鼠模型中CK1和CK10的表达明显上调且定位异常。 Objective: To investigate the expression of epidermal CK1 and CK10 in chronic ultraviolet irradiated mice. Methods: The mice were randomly divided into blank control and UV-damaged group, 20 mice per group. The model of chronic UV-damaged mice was established by ilTadiation with UVA plus UVB using simulated solar light. The irradiation began with UVB at 0.07 J/cm2, and UVA at 0.7 J/cm2 once daily, 5 days/week, followed by a weekly dose increase of 0,5 MED for 9 weeks. The total doses of UVB and UVA were 9.45 J/cm2 and 94.5 J/cm2, respectively. HE and Weigert staining were used to assess the cutaneous damage induced by UV irradiation. The expression levels of epidermal CK1 and CKI0 were evaluated with immunohistochemical staining before(W0) and after 9-week irradiation (W9). The paired t-test and independent sample t-test were used for statistical analysis. Results: The expression levels of CK1 and CK10 were similar between W0 and W9 in the blank control group (P〉0.05). In contrast, the expression levels of CK1 and CK10 were significantly different between W0 and W9 in chronic UV-damaged group (P〈0.05). Moreover, UV irradiation also altered the localization of CK1 and CK10 in the epidermis. Conclusion: Chronic UV irradiation upregulates the'expression of epidermal CK1 and CK10, and alters their localization.
