摘要
目的考察并优化流化床包衣法制备银耳多糖肠溶微粒(ETPP)工艺条件。方法以Eudragit L30-D55水分散体为包衣材料,柠檬酸三乙酯为增塑剂,滑石粉为抗黏剂,同时考察包衣后微粒的累计释放度,制备直径小于190μm肠溶微粒。结果载药量为40.62%,包封率为79.29%,酸中累计释放度小于10%,碱液中累计释放度大于75%,制备的微粒其平均粒径在115~190μm之间,圆整度、堆密度、总收率均较理想。结论应用流化床包衣法制备ETPP,其工艺简便,稳定可行,适合小鼠药物药理学和毒理学研究的需要和大工业生产。
Objective To review and optimize the process conditions of Enteric soluble microparticles of the polysaccharide from Tremella fuciform Berk( Tremella polysaccharide) by fluid-bed coating method. Methods With Eudragit L30-D55 as coating material,triethyl citrate as plasticizer,talcum powder as the antisticking agent,the release of the inspection after the coating particles respectively in degrees,preparation of diameter of less than 190 μm particles.Results 40. 62% drug loading. The encapsulation efficiency is 79. 29%,and the cumulative release of acid is less than 10%,the cumulative release of lye is greater than 75%. Preparation of particles of the average particle size is between 115 μm to 190 μm,its roundness,bulk density,in vitro release and total yield are ideal. Conclusion The methods of fluidized bed coating to prepare the enteric soluble microparticles of polysaccharides are simple,stable and feasible,and the methods are suitable for preparing the sample for the functional research of enteric administration with mouse and for industrial production.
