胃癌病人血清miR-338-3p的表达及其对胃癌细胞生物学行为的影响

摘要 目的:研究胃癌病人血清mi R-338-3p的表达及其对胃癌细胞生物学行为的影响。方法:提取40例胃癌病人及37例健康志愿者血清总mi RNA,采用逆转录实时荧光定量聚合酶链反应检测mi R-338-3p的表达。基于生物信息学预测其功能,采用细胞转染、侵袭、增殖实验及流式细胞术、Western印迹,研究其对胃癌细胞生物学行为的影响。结果:胃癌病人血清mi R-338-3p表达显著低于健康志愿者(P<0.001)。体外实验证明,转染mi R-338-3p后可明显抑制胃癌细胞株SGC7901和BGC823增殖(P<0.01)以及细胞侵袭能力,并促进其凋亡。同时抑制基质金属蛋白酶(matrix metalloproteinase,MMP)2蛋白的表达(P<0.05)。结论:胃癌病人血清mi R-338-3p表达明显降低。mi R-338-3p具有抑制胃癌细胞增殖和侵袭、促进凋亡的作用。可能通过下调MMP2表达抑制胃癌细胞。 Objective To investigate the expression level of serum mi R-338-3p of gastric cancer patients and its effect on the biological behavior of gastric cancer cells.Methods Total mi RNA in serum of 40 gastric cancer patients and 37 healthy controls was extracted.The expression level of mi R-338-3p in two groups was detected by quantitative real-time PCR.The analysis of bioinformatics was done to predict the function of targeted genes of mi R-338-3p which was further validated via cell biological techniques including cell transfection,proliferation and invasion experiments,flow cytometry and Western blot.Results The level of serum mi R-338-3p was significantly lower in gastric cancer patients compared with healthy control(P〈0.001).It was studied using gastric cells SGC7901 and BGC823 that mi R-338-3p significantly inhibited proliferation(P〈0.01) and invasion and prompted apoptosis.mi R-338-3p also significantly inhibited the expression level of matrix metalloproteinase(MMP) 2(P〈0.05).Conclusions It was shown in this study that the expression of serum mi R-338-3p decreased much in gastric cancer patients.mi R-338-3p could inhibit the proliferation and invasion of gastric cancer cells and induce the apoptosis.mi R-338-3p could inhibit the expression level of MMP2 in gastric cancer cells which may be the mechanism of biological behaviors of mi R-338-3p.
