摘要
目的:观察4种Calpain抑制剂不同浓度及作用时间对肝癌细胞MHCC97-H中Calpain-2蛋白表达的影响。方法:实验分为对照组、DMSO组及4种抑制剂组,4种抑制剂组分别给予高、中、低浓度处理肝癌MHCC97-H细胞24、48及72 h;用Western blot方法检测MHCC97-H中Calpain-2蛋白的表达,确定4种抑制剂的最适浓度和作用时间。结果:与对照组、DMSO组、其他时段各浓度组及同时段其他浓度组比较,高浓度ALM作用72 h后肝癌细胞MHCC97-H中Calpain-2蛋白的表达下降(P<0.05);与对照组、DMSO组、其他时段各浓度组及同时段其他浓度组比较,高浓度Calpain inhibitor IV、Calpain inhibitor VI、Calpastatin抑制剂72 h组肝癌细胞MHCC97-H中Calpain-2蛋白的表达量显著下降(P<0.01)。结论:4种高浓度Calpain抑制剂处理肝癌细胞MHCC97-H 72 h后可抑制肝癌细胞MHCC97-H中Calpain-2蛋白的表达。
Objective: To observe the effect of different concentration of Calpain inhibitor and different time on the expression of Calpain-2 protein in hepatocellular carcinoma cell line MHCC97-H.Methods: There were control group,DMSO group and 4 inhibiting group in the experiment. In the inhibiting groups,hepatocellular carcinoma MHCC97-H cells were treated with high,middle and low concentration of 4 different Calpain inhibitors for 24 h,48 h and 72 h,respectively. The expression of calpain-2 protein in MHCC97-H cells was detected by Western blot to determine the optimum concentration and time for the four inhibitors. Results: Compared with the control group,DMSO group,other groups of different concentration and different inhibiting time,the expression of Calpain-2 protein in HCC cell line MHCC97-H was significantly decreased after high concentration of ALLM inhibiting for72 h( P 〈0. 05). Compared with the control group,DMSO group,other groups of different concentration and different inhibiting time,the expression of Calpain-2 protein in HCC cell line MHCC97-H was significantly decreased after high concentration of Calpain inhibitor IV,Calpain inhibitor VI,and Calpastatin inhibitor inhibiting for 72 h( P 〈0. 01). Conclusion: The expression of Calpain-2 protein in hepatocellular carcinoma cell line MHCC97-H can be inhibited by 4 high concentration of Calpain inhibitors in the treatment of hepatocellular carcinoma cell line MHCC97-H 72 h.
