摘要
本试验利用分光光度法测定了关山樱花总黄酮含量,研究了关山樱花总黄酮对亚硝酸盐的清除作用。结果表明:关山樱花中总黄酮含量测定的最佳显色体系为Al(NO_3)_3-NaNO_2-Na OH显色体系,该方法最佳测定时间为显色后20~45 min范围内,测定方法精密度RSD=0.49%(n=5),加标回收率为102.55%(RSD=1.13%),利用该法测得关山樱花样品总黄酮含量为11.25%(RSD=1.44%,n=5);在体外模拟胃液条件下,关山樱花总黄酮浓度在4.00~96.00μg/m L范围内,对亚硝酸盐的清除率从14.64%±0.83%增加到99.42%±0.40%,半数清除率IC50值为18.85μg/m L,清除效果强于VC。该测定方法简便准确,且获得的关山樱花总黄酮对亚硝酸盐的清除效果显著。
The content of total flavonoids in flowers of Cerasus lannesiana Alborosea was determined by spectrophotometry. The scavenging action of total flavonoids on nitrite was studied. The results showed that the optimal chromogenic system was Al( NO_3)3- NaNO_2- Na OH,the optimal determination time was 20 ~ 45 min,the relative standard deviation( RSD) of precision degree was 0. 49%( n = 5),the adding standard recovery rate was 102. 55% with the RSD of 1. 13%,and the total flavonoid content was 11. 25% with the RSD of 1. 44%. Under the condition of simulated human gastric juice in vitro,the concentration of total flavonoids was 4. 00 ~ 96. 00 μg / m L,the scavenging rate increased from 14. 64 % ± 0. 83% to 99. 42% ± 0. 40%,the IC50 was 18. 85 μg / m L,and the scavenging effect was obvious and superior to VC. The method was simple and accurate.
