三种自噬调控蛋白在腮腺良、恶性多形性腺瘤中的表达及意义 被引量:4

摘要 目的:研究自噬调控中的主要蛋白雷帕霉素靶蛋白(mTOR)、蛋白激酶B(AKT)、Beclin 1在良、恶性多形性腺瘤中的表达及意义。方法:用免疫组织化学显色检测p-AKT和pmTOR在腮腺癌在多形性腺瘤、良性多形性腺瘤和正常腮腺组织中的表达。免疫印迹检测p-AKT、p-mTOR、Beclin 1在各组织中的表达。结果:p-AKT在正常腮腺组织、良性多形性腺瘤、癌在多形性腺瘤中的阳性表达分别为15%、40.5%、59.1%。p-mTOR在正常腮腺组织、良性多形性腺瘤、癌在多形性腺瘤中的阳性表达分别为20%、43.2%、65.7%。AKT、mTOR的表达在正常腮腺组织、良性多形性腺瘤、癌在多形性腺瘤中呈递增的趋势。Beclin 1在3种组织中呈递减趋势。结论:mTOR、AKT在大部分腮腺癌在多形性腺瘤组织中呈现过度表达而Beclin 1表达下降,这可能与腮腺癌在多形性腺瘤病因、病理机制相关。 Objective: To explore the expression and significance of mTOR, AKT and beclin 1 in parotid pleomorphic adenoma and carcinoma in pleomorphic adenoma. Methods: The expressions of p-ANT and p-roTOR in carcinoma in pleomorphic adenoma, parotid pleomorphic adenoma and normal parotid tissues were measured by immunohistochemical staining. The clinical and pathological factors were analyzed afterwards. The expression of beclin 1 was assessed by Western blotting. Results: The expression of p-ANT in normal parotid tissues, parotid pleomorphic adenoma and carcinoma in pleomorphic adenoma was 15 %, 40.5 %, 59.1% respectively and the expression of p-mTOR was 20%, 43.2%, 65.7% respectively. The expression of ANT/mTOR protein increased gradually in thee specimens. The positive rate of beclin 1 was descending in three tissues. Conclusion.. mTOR and AKT overexpress in most of carcinoma in pleomorphic adenoma. The positive rate of beclin 1 is descending. They may play an important role in occurrence and development of carcinoma in pleomorphic adenoma.
