摘要
目的:观察生川乌、生半夏与生川乌不同配伍比例对大鼠肾组织超微结构的影响。方法:SD大鼠,随机分为正常对照组、生川乌组、生川乌配不同生半夏组(1∶0.25组、1∶0.5组、1∶1组)。各给药组生川乌给药剂量均为0.8g/kg,连续灌胃4周,电镜下观察肾细胞超微结构变化。结果:生川乌组肾小管上皮胞质明显溶解,细胞膜内褶断裂,线粒体外膜破裂,嵴不规则或溶解;肾小球毛细血管内皮、线粒体肿胀,基膜不均,部分足突融合。生川乌配生半夏1∶0.25组肾小管上皮细胞质溶解略减轻,线粒体外膜断裂,嵴密集;肾小球毛细血管内皮轻度肿胀,基膜较均匀,部分足突融合。生川乌配生半夏1∶0.5组肾小管上皮细胞膜模糊,细胞质溶解减轻,细胞膜内褶、线粒体嵴仍有局部断裂或溶解现象;肾小球毛细血管内皮轻度肿胀,基膜较均匀,少量足突融合。生川乌配生半夏1∶1组肾小管上皮细胞质溶解明显减轻,细胞膜内褶多正常,内褶间线粒体排列不甚规则;肾小球毛细血管内皮肿胀、足突融合明显减轻,基膜厚度较均匀。结论:生川乌水浸液灌胃可导致大鼠肾超微结构损伤,生半夏与生川乌不同配伍比例水浸液灌胃可减轻损伤,其减轻程度随配伍比例增高而升高。
Objective: To explore the effect of raw Radix Aconite with different proportions of raw Penillia ternate on the ultrastructure of rat kidney. Methods: SD rats were randomly divided into five groups: normal control group, raw Radix Aconite group, raw Radix Aconite combining with raw Penillia ternate in the ratio of 1 : 0.25,1 : 0.5,and 1 : 1. 0. 8 g/kg raw Radix Aconite with different proportions of raw Penillia ternate were administered by gastric perfusion for 4 weeks, then observed the ultrastructure changes under transmission control group was normal. In the raw Radix Aconite electron microscope(TEM). Results: The ultrastructure of normal group, changes of the kidney tubule included obviously dissolved cytoplasm, folded and fractured plasma membrane, reduced organelles, ruptured mitoehondrial membrane, irregular and dissolved mitochondrial cristae; swollen glomerular capillary endothelial cells and mitochondria, basement membrane with non-uniform thickness and foot process fusion were also observed. Pathological changes of kidney tubule and glomerulus were lightened in raw Radix Aconite combining with raw Penillia ternate in the ratio of 1 " 0.25 and 1 : 0.5. The raw Radix Aconite combining with raw Penillia ternate in 1 : 1 group showed less pathological change under TEM. Compared with other drug administration groups, less cytoplasm was dissolved in renal tubular epithelial cell, most plasma membrane was normal except for some irregular mitochondrial alignment in the fractured zone; glomerular capillary endothelial cells were less swollen, thickness of basement membrane became uniform, and a little foot process was fuesd. Conclusion: Raw Radix Aconiti can cause obvious ultrastructure damage to rat kidney, but when combines with different proportions of raw Penillia ternate, damage decreases gradually with increasing proportion of Penillia ternate.
