活性维生素D_3对UUO模型大鼠肾间质骨形成蛋白-7表达的影响 被引量:1

摘要 目的:研究活性维生素D_3在肾小管间质纤维化病理过程中对骨形成蛋白-7(bone morphogenetic protein-7,BMP-7)表达的影响。方法:36只清洁级雄性Wistar大鼠,随机分为活性维生素D_3治疗组(n=12)、UUO模型组(n=12)、假手术组(n=12)。活性维生素D_3治疗组给予罗盖全(骨化三醇)灌胃,持续28d。假手术组及UUO模型组给予等量生理盐水灌胃。实验结束时,测定各组大鼠血清肌酐(Scr)和血尿素氮(BUN)值,肾脏病理检查造模侧肾小管间质纤维化程度,免疫组化法测定BMP-7在各组大鼠造模侧肾脏的表达。结果:1 UUO模型组的肾间质纤维化面积高于活性维生素D_3治疗组(P<0.05)。2BMP-7在假手术组明显高于UUO模型组和活性维生素D_3治疗组,而活性维生素D_3治疗组的BMP-7的表达高于UUO模型组(P<0.05)。结论:1BMP-7参与了肾间质纤维化的发生发展。2活性维生素D_3可以上调BMP-7的表达,抑制肾间质纤维化。 Objective: To observe the expression and distribution of the Bone morphogenetic protein-7( BMP-7) in kidney tissues in the process of renal interstitial fibrosis; to explore the influence of active vitamin D_3 on renal tubule interstitial fibrosis and on the expression BMP-7 in UUO model rat. Methods: 36 SD rats were randomly divided into active vitamin D_3 treatment group( n = 12),UUO model group( n = 12),sham operated group( n = 12). The expression and distribution of the BMP-7 in the kidney tissues were detected by immunohistochemistry. Results: The expression of BMP-7 decreased in UUO model group and in active vitamin D_3 treatment group compared with the tofsham operated group. The expression of BMP-7 in UUO model group was lower than that in the active vitamin D_3 treatment group( P〈0. 05). Conclusion: 1BMP-7 take part in renal interstitial fibrosis.2Active vitamin D_3 can increase the expression of BMP-7 and relieve renal interstitial fibrosis.
