心肌尔康对L-NAME诱导高血压小鼠的降压作用和对靶器官损伤的影响 被引量:1

摘要 目的探讨心肌尔康对一氧化氮合酶抑制剂L-NAME诱导高血压小鼠心血管重构的影响。方法实验分为正常组、模型组、心肌尔康组。每周测小鼠尾动脉压,实验第8周末,处死小鼠,计算小鼠心脏指数、肺重指数,HE染色和V-G染色观察心脏、胸主动脉病理形态学改变。测定小鼠血浆NO含量。结果与正常组相比,模型组血压明显升高(P<0.01),心脏指数、心肌细胞横截面积、心肌纤维化明显升高,胸主动脉发生明显重构。血浆NO含量降低;与模型组相比,心肌尔康能明显降低L-NAME诱导的高血压小鼠的血压(P<0.01),升高NO含量,改善心血管重构。结论心肌尔康能明显抑制L-NAME诱导小鼠血压的升高,改善心血管重构。 Objective To investigate the effects of Xinjierkang(XJEK)on L-NAME induced hypertension and cardiovascular remode-ling in mice.Methods Mice were assigned into control group,model group,and Xinjierkang group,At the end of 8^th week,the mice were euthanatized and the heart,lung and thoracic aorta were harvested,The heart weight/body weight(HWI),lung weight/body weight (LWI)were calculated,respectively,HE staining and /or V-G staining were used to detect cardiac and thoracic aortic pathological changes,Colorimetric analysis was used to assay nitric oxide(NO)in plasma.Results Compared with control group,the systolic blood pressure,HWI,LWI,cross-sectional area of cardiomyocytes,myocardial fibrosis were significantly increased (P 〈0.01 ),as well as marked thoracic aortic remodeling,decreased plasma NO content,Administration with XJEK markedly inhibited the elevation of systolic blood pressure,HWI and LWI,which blunted the decrease of NO plasma content,ameliorated cardiovascular remodeling.Conclusions XJEK has protective effects against cardiovascular remodeling in L-NAME induced hypertensive mice.
