不同发育期大通牦牛组织线粒体COX基因的分析 被引量:1

摘要 采用绝对定量荧光PCR(Real-Time PCR),对妊娠4月、出生1日、1月、3月和6月龄不同发育期的青海大通牦牛组织中线粒体COX-Ⅰ及COX-Ⅱ的mRNA表达量进行分析。结果显示,不同发育期牦牛大脑、心肌、骨骼肌、肝组织线粒体COX-ⅡmRNA表达量均高于COX-ⅠmRNA表达量;COX-ⅡmRNA表达量与COX-Ⅰ相比较稳定。妊娠4月龄大脑组织线粒体COX-Ⅰ、ⅡmRNA表达量均显著高于其他年龄组(P<0.05);1日龄心肌组织线粒体COX-Ⅰ、ⅡmRNA表达量均显著高于其他年龄组(P<0.05);1月龄骨骼肌组织线粒体COX-Ⅰ、ⅡmRNA表达量均显著高于其他年龄组(P<0.05);6月龄肝组织线粒体COX-ⅠmRNA表达量最高,差异显著(P<0.05);而肝组织线粒体中COX-ⅡmRNA表达量在各个年龄段差异不显著(P>0.05)。结果表明,大通牦牛生长发育阶段妊娠4月龄胎儿大脑组织、出生1日龄心肌组织及1月龄骨骼肌组织COX亚基mRNA转录水平均表现为上调表达,这与组织器官处于快速发育阶段及与其发挥重要的功能有直接关系。 Qinghai Datong yak was regarded as the research object and the absolute quantitative Real-time PCR was performed to analyze the expression level of COX-Ⅰ COX-Ⅱ mRNA in the mitochondria of tissues at different developmental stages(pregnancy for 4 months;1 day,1 month,3 months and 6months after birth,respectively).Results indicated that the expression level of COX-Ⅱ mRNA in the mitochondrial of cardiac muscle,skeletal muscle,brain,and liver was significantly higher than that of COX-ⅠmRNA.Compared with the expression level of COX-ⅠmRNA in different tissues,COX-Ⅱ mRNA was relatively stable.For the expression level of COX-Ⅰand COX-Ⅱ mRNA in the mitochondrial of brain,pregnancy for 4 months group was significantly higher than other groups(P〈0.05).For the expression level of COX-Ⅰand COX-Ⅱ mRNA in the mitochondria of cardiac muscle,one-day-old group was significantly higher than other groups(P〈0.05).For the expression level of COX-Ⅰand COX-Ⅱ mRNA in the mitochondria of skeletal muscle,one-month-old group was significantly higher than other groups(P〈0.05).For the expres sion level of COX-ⅠmRNA in the mitochondria of liver,6-month-old group was significantly higher than other groups(P〈0.05),while COX-Ⅱ mRNA expression level of different age group in the mitochondria of liver was not significantly different(P〉0.05).In conclusion,transcript level of Datong yak COX-Ⅱ mRNA in different stages of growth and development(pregnancy for 4 months in brain;1day after birth in cardiac muscle and 1 month after birth in skeletal muscle respectively) was up-regulated,which had a direct relationship with tissues in rapid developmental stage as well as their important function.
